目的检测E381基因在脊髓运动神经元(SMN)轴突生长抑制前后的表达变化及意义。方法应用逆转录-聚合酶链反应(1iT-PCR)和Westernblot检测SMN轴突生长抑制前后E381mRNA和蛋白表达的变化,应用免疫细胞化学检测培养SMNⅢ型β-微管蛋白的表达。结果与正常培养的SMN比较,与轴突抑制物共培养的SMNⅢ型β-微管蛋白表达明显降低(P〈0.01),E381 mRNA和蛋白的表达均显著升高(P〈0.01)。结论E381基因的过度表达可能是SMN轴突生长抑制的重要因素。
Objective To investigate the expression of E3B1 gene in clultured spinal motoneuron (SMN) and its implication. Methods The mRNA and protein levels of E3B1 were detected by RT-PCR and Western blot respectively in the cultured neurons. Results The mRNA and protein levels of E3B1 in the neurons with axonal growth restricted were obviously higher than those in the normal cultured neurons (P 〈0.01 ). Conclusion The expression changes of E3B1 gene has a intrick relationship with neurite outgrowth.
