目的:观察哮喘大鼠骨形态构建蛋白2,4(BMP-2,BMP-4)表达情况。方法:SD大鼠45只,随机分为2组,对照组(Ⅰ组)为健康大鼠;哮喘模型组(Ⅱ组)为卵清白蛋白(OVA)致敏后吸入激发建立的模型。HE染色后图像分析检测大鼠气道形态学参数,免疫组化方法用于BMP-2,BMP.4表达定位,Western蛋白印迹法测定BMP-2,BMP-4含量。结果:Ⅱ组与Ⅰ组相比,支气管内径缩小。平滑肌层增厚(P〈0.05);BMP-2表达增加12.3%。BMP-4表达减少7.9%(P〈0.05)。结论:哮喘大鼠肺内BMP-2,BMP-4可能参与哮喘病理生理变化调控。
Objective: To investigate the expressions of bone morphogenetic protein-2 (BMP-2) and BMP-4 in the lungs of rats with asthma. Methods: Forty-five SD rats were randomly divided into2 groups: group 1 (control group) and group 2 (asthma group). The asthmatic rat model was established by ovalbumin (OVA) sensitization and challenge. The changes of airway, the expressions of BMP- 2 and BMP-4, and the contents of BMP-2 and BMP-4 were determined by using image analysis system,immunohistochemical method, and Western Blotting, respectively. Results: Compared with group 1, the bronchial internal diameter decreased and the thickness of bronchial smooth mascle increased in group 2 ( P 〈 0.05 ). The expression of BMP-2 was significantly higher than that in group 1 ( 42. 1 ± 6.2vs. 29.8 ± 8.4% , P 〈 0.05 ) , and the expression of BMP-4 in group2 was signiificantlylower than that in group 1 (11.5 ±4.2 vs. 19.4 ±5.3%, P 〈0.05). Conclusion: The increase in BMP-2 expression and the decrease in BMP-4 expression may participate in the pathogenesis of asthma in rats.
