由于抗生素药物的不合理使用等原因,病原微生物耐药性问题日益突出,严重影响了细菌性感染疾病的治疗效果,也逐渐成为目前人类着重关注的健康问题。微生物可通过多种方式来抵御抗生素引起的毒性,其中利用耐药泵将药物直接外排的方式是细菌主要的耐药性机制之一。通过对耐药泵结构和机制的研究,有利于开发以多药耐药泵为作用靶点的细菌敏感型药物,对于降低细菌耐药性和进行临床治疗有重要指导意义。文章概述了当前细菌的多药耐药外排泵的主要分类和分布、结构特征、作用机制及表达调控等方面,阐明了多药耐药外排泵抑制剂的研究意义和价值,并对其研究前景进行展望。
During the past decades, the drug resistant pathogenic microorganism has been an increasingly serious problem with the abuse of antibiotics which have threatened the effective treatment of bacteria infections. Consequently, it also has caused people's high attention and continued to be a global concern. It is shown that the microorganism has many ways to resist the antibiotics and the mutidrug-restistance efflux pump is one of the most important paths. Recently,the study is focusing on how to inhibit the activity of mutidrug-restistance efflux pumps. It's quite helpful to develop new bacterial sensitive drugs using the efflux pumps as target through the structure and molecular mechanism study. The research will do a great help to slow down the drug resistance of bacterial in clini- cal therapy. In this paper, the main distribution, classification, structure features, mechanism of action and regulation of expression are summarized. The research significance and value as well as the research prospects are also clarified.
