目的:观察隔药饼灸对动脉粥样硬化(AS)兔主动脉内皮细胞核因子-κB(NF-κB)及细胞间黏附分子-1(ICAM-1)表达的影响,探讨隔药饼灸干预AS形成的作用机制。方法:将50只新西兰大耳白兔随机分为5组:空白组、模型组、直接灸组、隔药饼灸组、西药组,每组10只,除空白组外,其余4组通过高脂饲料喂养及免疫损伤法建立兔AS模型。分别用RT-PCR技术、荧光定量PCR技术检测兔主动脉内皮细胞NF-κB mRNA、ICAM-1 mRNA表达。结果:与空白组相比,模型组主动脉内皮细胞NF-κB mRNA、ICAM-1 mRNA表达差异具有统计学意义(P〈0.01);与西药组相比,隔药饼灸组NF-κB mRNA、ICAM-1 mRNA表达差异无统计学意义;直接灸组NF-κBmRNA、ICAM-1 mRNA表达差异有统计学意义(P〈0.05)。结论:隔药饼灸和直接灸抗AS与其降低AS兔主动脉内皮细胞NF-κB mRNA、ICAM-1 mRNA表达有关,隔药饼灸作用优于直接灸。
Objective: To observe the effects of Cake-separated Moxibustion on the expression of nuclear factor-kappa B(NF-κB) and lintercellular adhesion molecule-1(ICAM-1) in aortic endothelial cells of atherosclerotic rabbits and explore whether Cake-separated Moxibustion could delay atherogenesis formation.Methods: 50 New Zealand white rabbits were randomly divided into 5 groups: blank group,model group,direct moxibustion group,Cake-separated Moxibustion group and western medicine group,10 rabbits in each group.Beside of blank group,other groups were established a rabbit model of atherosclerosis by using immunologic injury combined with hypercholesterol diet.NF-κB mRNA in aortic endothelial cells were measured by reverse transcription polymerase chain reaction(RT-PCR) and ICAM-1 mRNA were detected by fluorescent quantitative PCR.Results: Compared with blank group,the expression of NF-κB mRNA and ICAM-1 mRNA in model group had statistical significance(P0.01);compared with western group,the expression of NF-κB mRNA and ICAM-1 mRNA in Cake-separated Moxibustion group had not significance;compared with Cake-separated Moxibustion group,the expression of NF-κB mRNA and ICAM-1 mRNA in direct group had statistical significance(P0.05).Conclusion: Cake-separated Moxibustion and direct moxibustion could all delay the formation of atherosclerotic plaque by decreasing the expression of NF-κB mRNA,ICAM-1 mRNA in aortic endothelial cells of atherosclerotic rabbits,while the effects of Cake-separated Moxibustion were more significant than direct moxibustion.
