以蛹虫草子实体为材料,经水提醇沉后获得沉淀样品P和上清样品L,初步抗HIV-1蛋白酶检测显示活性成份集中在样品L中.进一步利用大孔吸附树脂和高压液相对L进行分离纯化,并对纯化样品的体外抗HIV-1蛋白酶活性和抗HIV-1逆转录酶活性进行了测定.结果显示,L经大孔吸附树脂和高压液相分离纯化后得到三种物质均具有抗HIV-1蛋白酶活性f1,f2和f3,其中f1对HIV-1蛋白酶抑制作用最强,比L提高了约29%;三种物质对HIV-1逆转录酶均有一定的抑制作用.
Cordyceps militaris is one of Chinese traditional medicines with activities of antioxidant and anti-tumor. In the present study, the aqueous extract of fruiting body from Cordyceps militaris was precipitated with ethanol. Then the precipitate (P) and the filtrate (L) were obtained. Anti-HIV-1 protease test showed that L was the main fraction with anti-HIV-1 protease activity. Therefore L was purified by using resin and HPLC. The anti-HIV activities of purified components were assayed in two different methods, the inhibition of HIV-1 reverse transcriptase and HIV-1 protease. These results demonstrated that three compounds, named as fl, f2 and f3, were obtained from L. Among them fl showed the strongest inhibitory protease activities and about 29 % higher than L; all of them had inhibitory to HIV-1 reverse transcriptase after purification.
