目的观察汕阿片受体基因内含子2的两个单核苷酸多态位点(rs9479757和rs2075572)与海洛因成瘾的相关性。方法用TaqMan荧光法检测332例海洛因成瘾者和190例健康对照者的rs9479757(31A/G)和rs2075572(691C/G)的基因型并进行关联分析。结果rs9479757和rs2075572在对照组和成瘾组间的多态分布差异无统计学意义(P〉0.05);rs9479757在海洛因成瘾轻型、中型及重型组间多态分布差异有统计学意义(P〈0.01);海洛因烫吸者rs9479757为GG基因型者的日均使用量,显著高于AG基因型者(P〈0.05)。结论μ阿片受体基因内含子2多态性与海洛因成瘾无明显相关性,但与海洛因成瘾者的严重程度及某些行为特征有关联。
Objective To explore the association between the single nucleotide polymorphism (SNP) in intron 2 of the I-II opioid receptor gene (OPRM1) and heroin addiction. Methods The genotypes of the two SNPs (rs9479757, rs2075572) in 332 heroin dependent patients and 190 healthy individuals( as control group) in male Han Chinese population were detected by TaqMan SNP genotyping technique, and the association between heroin dependence and the two SNPs was analyzed. Re- suits The frequencies of genotype and allele at rs9479757, rs2075572 were not significantly different between the heroin addicts and the control groups (P 〉 0. 05 ). The frequencies of genotype and allele at rs9479757 in the mild, moderate and heavy clusters of heroin addicts were signifi- candy different (P 〈 0. 01 ). Furthermore, in the smoking group, the average heroin daily dosages of GG genotype at rs9479757 was higher than that of AG genotype ( P 〈 0. 05 ). Conclusion The polymorphisms (rs9479757, rs2075572 ) in intron 2 of the μ opioid receptor gene (OPRM1) may not play the major role in heroin dependence, but have certain association with some behavioral characters of heroin addicts.
