根据香菇单核菌丝体135A 和135B 的全基因组测序,通过生物信息学方法分析筛选出香菇功能基因 CAP、GLA1、TLG1结构域中的非同义 SNP 位点,开发出3对 SNP-CAPS 分子标记。结果表明:这3对功能基因SNP-CAPS 分子标记可将23个香菇单核体供试菌株分成135A 型(分别为16株、20株、2株)和135B 型(分别为7株、3株、21株),这些标记表现出的多态性为进一步研究香菇菌株的来源和功能基因的定位提供参考价值;并建立起可以简单、快速、准确地调查重要功能基因的 SNP 位点变异情况的方法。
Based on the whole genome sequencing of monokaryon strain of Lentinula edodes 135A and 135B,the non-synonymous SNP loci within the domain of the CAP,GLA1,TLG1 genes were found by using the bioinformatics way,and developed into three pairs of primers as SNP-CAPS molecular markers.The results showed that these SNP-CAPS markers could divide 23 tested monokaryons strains of L.edodes into two groups:135A type(corresponding 16,20 and 2 strains)and 135B type(corresponding 7,3 and 21 strains)which estab-lished the method to detect the distribution of known SNP loci in the population of L.edodes.This method will be helpful for tracking the source of L.edodes strains and identifying the functional geneseasily,quickly,and accu-rately.
