目的 探讨美洲大蠊提取物体外对肝癌(HCC)HepG_2细胞增殖、迁移能力,以及细胞中血管内皮生长因子(VEGF)蛋白表达的影响。方法 噻唑蓝(MTT)法检测细胞增殖;划痕实验检测美洲大蠊提取物对HCC细胞迁移能力的影响;免疫细胞染色法、酶联免疫吸附试验(ELISA)检测美洲大蠊提取物对细胞中VEGF蛋白表达的影响。结果 MTT和划痕实验结果表明,美洲大蠊多肽、CⅡ-3和脱脂膏能抑制人HCC HepG_2细胞的增殖和迁移,且呈时间、剂量依赖关系;免疫细胞化学染色法和ELISA法结果显示,美洲大蠊多肽、CⅡ-3及脱脂膏能下调人HCC HepG_2细胞中VEGF蛋白的表达。结论 美洲大蠊多肽、CⅡ-3及脱脂膏能抑制人HCC HepG_2细胞增殖和迁移,同时能降低HepG_2细胞中VEGF蛋白的表达,且美洲大蠊多肽的作用优于CⅡ-3和脱脂膏。
Objective To investigate the effect of extracts from Periplaneta Aamericana on in vitro prolife- ration, migration and VEGF protein expression of human hepatocellular carcinoma HepG2 cells. Methods Scratch assay was applied to measure the effect of extracts from Periplaneta americana on migration of HepG2 cells. Immunohistochemical staining and enzyme-linked immunosorbent assay (ELISA) were used to detect the effect of extracts from Periplaneta america on the expression of VEGF protein in the cells. Results MTI" assay and Scratch assay showed that Periplaneta americana polypeptides, CⅡ-3and skimmed cream could inhibit the proliferation and migration of HepG2 cells in a time- and dose-dependent manner. Immunohistochemical staining and ELISA indicated that Periplaneta americana polypeptides,CⅡ-3 and skimmed cream could down regulate the expression of VEGF protein in the cells. Conclusions Periplaneta americana polypeptides, CⅡ-3 and skimmed cream can inhibit the in vitro proliferation and migration of HepG2 cells in vitro in a time- and dose-dependent way, and reduce the expression of VEGF protein in the cells; and the effect of Periplaneta americana extracts is better than that of CⅡ-3 and skimmed cream.
