目的:研究针刺治疗骨骼肌过度使用损伤的疗效。方法:18只雄性SD大鼠随机分成安静对照组(C)、1周自然恢复组(1RG)和1周针刺恢复组(1RAG)3组。通过4周低强度离心运动方式建立过度使用损伤动物模型(动物跑台、一160、60.90分钟/天,5天/周)。恢复1周后,在第5周取股中肌。采用透射电子显微镜观察骨骼肌细胞超微结构损伤累积;采用扫描电子显微镜观察骨骼肌神经、血管结构损伤和胶原纤维结构变化;采用免疫组化方法量化沉积的胶原纤维中collagenI纤维蛋白浓度。结果:l周自然恢复组呈现过度使用损伤,包括Z线水波纹样变化,肌原纤维间隙变宽,肌原纤维降解.肌膜下线粒体聚积,肌纤维内线粒体降解物聚积,肌肉内神经和血管纤维化,以及肌内膜尤其是肌束膜厚度增加、collagenI纤维蛋白浓度增加。针刺显著减轻了骨骼肌过度使用损伤(P〈0.01)。包括肌原纤维降解减少,骨骼肌超微结构损伤修复,线粒体数量增加,骨骼肌神经和血管结构损伤减弱.以及肌内膜尤其是肌束膜的厚度减少、collagenI纤维蛋白浓度下调。结论:针刺能治疗离心运动引起的骨骼肌过度使用损伤,其效应-9线粒体数量增加有关。
Objective The aim of this study was to investigate the effects of acupuncture therapy on the eccentric exercise-induced skeletal muscle overuse injuries. Methods Eighteen male Sprague- Dawley rats were assigned randomly to three groups :the control group (C), 1 week recovery group (1 RG)and 1 week received intramuscular acupuncture recovery group (1 RAG). Animal model of overuse injury was experimentally induced through 4-week low-intensity eccentric exercise (treadmill running, -16~ ,60 ~90 min/day,5 days/week). After 1-week recovery,samples of vastus intermedius were collected at the 5th week. Skeletal muscle cells uhrastructural damage,and nerves,blood vessels structural damage , protein levels of collagen I in depositive collagen fiber, and collagen structural change of skeletal muscle were observed. Results After 4-week repetitive eccentric exercise,skeletal muscle overuse injuries,including Z-band streaming,widened intermyofibrillar spaces,myofibril degradation, aggregated subsarcolemmal mitochondria,fibrosis of nerves and blood vessels,thickened endomysium (especially perimysium),and increased collagen [ protein levels in 1 RG appeared. Needle puncture weakened the overuse injuries in 1 RAG as compared to that in 1 RG (P 〈 0.01),including decreased myofibril degradation,restoration of uhrastructural damage in skeletal muscle cells,increase in the numbers of mitochondria,alleviated nerves and blood vessels structural damage,and decreased collagen I protein levels in expanded endomysium,particularly in perimysium. Conclusions Acupuncture could effectively treat eccentric exercise-induced skeletal muscle overuse injuries. The effectiveness of acupuncture seems to be associated with increased numbers of mitochondria in skeletal muscle.
