目的:以药用真菌桑黄为研究对象,应用甲醇为溶剂提取其有效成分,进一步采用微孔读板法评价桑黄提取物的抗氧化活性。方法:应用多功能读板器,在已有抗氧化活性评价方法的基础上进行适当修改,以满足微量检测的需要,采用96-微孔板法快速评价不同浓度的桑黄提取物对羟基自由基、超氧阴离子自由基和DPPH自由基的清除能力。结果:通过微孔读板法快速评价桑黄提取物抗氧化活性的结果表明,桑黄提取物对三种自由基(·OH、O2-·和DPPH自由基)均有一定的清除能力,并且不同桑黄提取物对自由基清除能力与其浓度存在量效关系。结论:采用修改后适合微量检测抗氧化活性评价方法,应用微孔读板法能够快速评价具有自由基清除活性的物质,该方法快速可行,适合高通量筛选和评价具有抗氧化活性的成分。
Objective:To evaluate the antioxidant activity of the methanol extract from Phellinus igniarius. Methods:Molecular Devices with 96-microplate was applied to evaluate the Phellinus igniarius extract capacity of scave nging hydroxyl radical( ·OH),superoxide anion radical(O2-·)and 1,1-diphenyl-2-picrylhydrazyl(DPPH)free radical via minor modification in this investigation. Results :The extract of Phellinus igniarius presents great scavenging capacity for ·OH,O2-· and DPPH radicals by Molecular Devices;and the efficiency increased with the content of extract from Phellinus igniarius. Conclusion :Molecular Devices was a feasible,effective,fast and high-throughput method for screening and evaluating the antioxidant activity of the crude extract.
