目的:探讨miR-455在宫颈癌中的表达及其与微血管密度(MVD)及血管内皮生长因子(VEGF)表达的关系.方法:采用实时荧光定量PCR技术检测68例宫颈癌患者和49例慢性宫颈炎(对照组)组织中miR-455的表达.免疫组化方法检测组织内MVD及VEGF蛋白表达水平.结果:宫颈癌组织内miR-455的相对表达量显著低于慢性宫颈炎(1.17 ±0.59 vs 2.58±0.83,P<0.01).68例宫颈癌标本MVD表达的计数结果为30.45 ±7.9.Pearson相关分析结果显示,宫颈癌组织内miR-455表达水平与MVD呈负相关(r=-0.835,P<0.01.宫颈癌VEGF表达为阴性、弱阳性、中度阳性和强阳性组中,miR-455相对表达定量(RQ)分别为1.77 ±0.58、1.59±0.50、1.34 ±0.35和0.68 ±0.26,Spearman等级相关分析结果显示,miR-455表达水平与VEGF的表达呈负等级相关(rs=-0.857,P<0.01).结论:miR-455低表达可能通过VEGF信号通路促进宫颈癌内微血管形成,是一个潜在的抑癌miRNA.
Objective:To investigate the expression of miR-455 in cervical cancer and its relationship with microvascular density(MVD) and vascular endothelial growth factor (VEGF).Methods:The expression of miR-455 in 68 cases with cervical cancer and 49 cases with chronic cervicitis(control group) was tested by real time PCR.The expression of MVD and VEGF were examined by immunohistochemistry.Results:The relative quantity of miR-455 expression in cervical cancer tissue was significantly lower than that in chronic cervicitis (1.17 ±0.59 vs 2.58 ±0.83,P < 0.01).The result of MVD was 30.45 ± 7.9 in 68 cases of cervical cancer,and the expression of miR-455 was negatively correlated with MVD by Pearson correlation analysis (r =-0.835,P < 0.01).The expression of VEGF was negative,weakly positive,moderately positive,and strongly positive in the specimen of cervical cancer,and the expression of miR-455 was 1.77 ± 0.58,1.59 ± 0.50,1.34 ± 0.35 and 0.68 ± 0.26 respectively.Spearman rank correlation analysis showed that the expression of miR-455 was negatively correlated with that of VEGF (rs =-0.857,P < 0.01).Conclusion:Low expression of miR-455 promotes the formation of microvascular in cervical cancer,and it may be a potential tumor suppressor gene.
