丙泊酚麻醉下大鼠反复腰椎穿刺鞘内给药的可行性研究

目的探讨骨癌痛大鼠脊髓背角蛋白激酶Cγ(PKCγ)表达的变化。方法成年雌性Wistar大鼠48只,随机分为3组(n=16):对照组、假手术组和骨癌痛组。对照组不给予任何处理,骨癌痛组右侧胫骨上部骨髓腔内注射Walker256乳腺癌细胞10μl制作骨癌痛模型;假手术组同部位注射热灭活的Walker256乳腺癌细胞10μl。各组于术前1d、术后3、6、9、12、15、18和21d时测定术侧后爪机械痛阈,并于术前1d、术后6、15和21d时各处死4只大鼠,取L4-6脊髓组织,采用免疫组化法测定脊髓背角PKCγ的表达水平。结果与术前1d时比较,骨癌痛组术后PKCγ表达上调,术后15d时达最大值(P〈0.05);与对照组和假手术组比较,骨癌痛组术后机械痛阈降低,PKCγ表达上调(P〈0.05);PKCγ表达水平与机械痛阂呈负相关(r=-0.984,P〈0.05)。结论骨癌痛大鼠脊髓背角PKCγ表达上调,该变化可能参与了骨癌痛的维持。

英文摘要:

Objective To investigate the changes in expression of protein kinase Cγ (PKCT) in the spinal dorsal horn in a rat model of bone cancer pain (BCP). Methods Forty-eight female adult Wistar rats weighing 180-200 g were randomly divided into 3 groups (n = 16 each): group I control (group C); group II sham operation (group S) and group III. BCP. In group BCP Walker 256 mammary gland cancer cell suspension 10 μl was injected into left tibia, while in group S heat-inactivated Walker 256 mammary gland cancer cell suspension 10 μl was injected into the same region. Mechanical pain threshold of hind paw was measured every 3 d from 1 d before operation to 21 d after operation, and then the rats were killed at 1 d before and 6, 15 and 21 d after operation (4 rats at each time point) and the L4.6 segment was removed for determination of PKCγ expression ( by immuno-histochemistry). Results PKCγ expression was up-regulated after operation and reached the highest value at 15 d after operation as compared with that at 1 d before operation in group BCP ( P 〈 0.05) . Mechanical pain threshold was significantly decreased and PKCγ expression was up-regulated in group BCP as compared with group C and S ( P 〈 0.05) . PKCγ expression was negatively correlated with mechanical pain threshold ( r = - 0. 984, P 〈 0.05). Conclusion The expression of PKCγ is up-regulated in spinal dorsal horn in the rat with BCP, the up-regulation of PKGγ expression may be involved in the maitenance of BCP.
