亚麻显性雄性核不育基因对亚麻育种以及杂种优势的利用具有相当重要的作用,为更好地利用这一优良生物性状,很有必要对其相关基因进行标记、克隆和序列分析。通过使用252条10-mer的随机引物对遗传背景相似的兄妹杂交分离出的17对可育株和不育株亚麻进行RAPD分子标记,得到三条差异片段,通过回收、克隆及测序,然后用BLAST进行序列分析以及对比研究,发现这3条差异片段与水稻不育基因具有同源性,因此推测其与亚麻显性雄性核不育有关。
Dominant male sterile gene of flax is very important for flax breeding and using of heterosis. For better utilization of these genes, It is necessary to marker and clone and sequence these genes. Three specific DNA fragments were gained by RAPD marker with 252 arbitrary 10-mer primers to identify dominant sterile gene of 17 pair of sterile and fertile flaxes breeding from parents with the same genetic background. Clone and Sequence as well as comparison of the fragments showed that the fragments is homology to sterile gene of rice. it is then assumed that the fragments are associated with flax dominant male sterile.
