结合天津市高校创新团队承担的国家自然科学基金项目,以更新生物技术及其相关专业综合实验项目为切入点,以节旋藻TJSD091藻株基因组的CRISPR位点引物为例,阐述了如何通过Touchdown PCR与梯度PCR技术结合确定PCR扩增最佳退火温度这一实验,激发学生的探究心理和学习兴趣,引导学生主动学习的做法,分析了将科研与实验教学相结合的优越性,认为该教学模式不仅培养了学生的科研思维、提高了学生解决实际问题的能力,而且可为应用型高校本科生实践教学体系的完善提供新思路。
This experiment is combined with the project of National Natural Science Foundation of innovation team in College of Tianjin City,to update the biotechnology and related professional's comprehensive experimental project as the breakthrough point,to the primer of CRISPR locus in the genome of Arthrospira sp. Taking TJSD091 as an example,the paper elaborates how to determine the optimal annealing temperature in PCR amplification reaction by the combination of Touchdown PCR and gradient PCR. The reform stimulates students 'inquiry psychology and study interests,guides students to take the initiative to learn. The advantages of combining scientific research with experimental teaching are analyzed,it is believed that this teaching mode can train students' scientific thinking,improve students' ability to solve practical problems, and can provide a new way for the improvement of the practical teaching system for the undergraduates in applied colleges.
