为进一步阐明大蓟的化学成分,运用硅胶、大孔吸附树脂(Diaion-101)、Sephadex LH-20等柱色谱和薄层色谱技术对大蓟进行提取分离,根据波谱数据和参考文献等手段从中鉴定了9个木脂素类化合物,分别为:爵床脂素B(1),6'-羟基爵床脂素A(2),6'-羟基爵床脂素B(3),山荷叶素(4),爵床脂素A(5),6-羟基-4-(4-甲氧基-3-甲氧基苯基)-3-羟甲基-5-甲氧基-3,4-二氢(3R,4S)-2-醛基萘(6),6-羟基-4-(4-甲氧基-3-甲氧基苯基)-3-羟甲基-7-甲氧基-3,4-二氢(3R,4S)-2-醛基萘(7),4'-去甲基鬼臼毒素(8),鬼臼毒素(9).化合物(1~9)均为首次从大蓟中分离得到.同时,本研究用D-氨基半乳糖对HL-7702细胞诱导产生毒性,采用MTT法对化合物(1~9)进行肝保护活性筛选,结果表明化合物2,3,4,7和8(10μm)具有一定的肝保护活性.
To investigate the chemical constituents of Cirsium japonicum,nine lignans were obtained by column chromatography on silica gel,macroporous resin( Diaion-101),Sephadex LH-20,and TLC for separation and purification. Their structures were identified as justicidin B( 1),6'-hydroxy justicidin A( 2),6'-hydroxy justicidin B( 3),diphyllin( 4),justicidin A( 5),6-hydroxy-4-( 4-methoxyl-3- methoxyphenyl)-3-hydroxymethyl-5-methoxyl-3,4-dihydro( 3R,4S)-2-aldehydenaphthalene( 6),6-hydroxy-4-( 4-methoxyl-3-methoxyphenyl)-3-hydroxymethyl-7- methoxyl-3,4-dihydro( 3R,4S)-2-aldehydenaphthalene( 7),4'-demethyl podophyllotoxin( 8),podophyllotoxin( 9) by spectroscopic data and references. Compounds( 1 ~ 9) were isolated from this plant for the first time. Meanwhile,Compounds( 1 ~ 9) were evaluated for their hepatoprotective activities against D-galactosamine-induced toxicity in HL-7702 cells using a MTT colorimetric method.
