目的:研究高糖诱导下,杭白菊黄酮对人脐静脉血管内皮细胞(HUVEC)表达Bax、Bcl-2的影响。方法:将HUVEC分为空白对照组、正常血糖组、高糖刺激组和杭白菊高、中、低剂量组,作用24和48h后,观察各组细胞形态变化,采用MTT法检测细胞生长状况,RT-PCR法检测内皮细胞Bax、Bcl-2的表达。结果:高糖诱导下,杭白菊黄酮可下调Bax的表达,上调Bcl-2的表达,而杭白菊黄酮呈剂量依赖性抑制Bax表达,增强Bcl-2表达。结论:杭白菊黄酮可通过下调高糖诱导的血管内皮细胞Bax表达,上调Bcl-2表达,抑制血管内皮细胞凋亡,在一定程度上有助于防治糖尿病血管病变,为临床应用杭白菊防治糖尿病患者血管并发症提供理论参考。
Objective: To study the effects of flavonoids in Chrysanthemum morifolium on the expression of Bax and Bcl-2 in human umbilical vein endothelial cells(HUVECs) induced by high sugar. Methods: The HUVECs were divided into a blank control group, a normal blood sugar group, a high, medium, low dosage group of chrysanthemum morifolium. The cells morphologic change in each group was observed, MTT method was used to examine the growth condition of cells, RT-PCR was used to detect the expression of Bax and Bcl-2 in vascular endothelial cell after 24 and 48 hours. Results: The induction of high sugar declined the expression of Bax and raised the expression of Bcl-2. And the flavonoids in Chrysanthemum morifolium was dependently correlated with its dosage to restrain the expression of Bax and enhance the expression of Bcl-2. Conclusion: Through the down-regulation of the expression of Bax in vascular endothelial cell and the up-regulation of the expression of Bcl-2 by the induction of high sugar, the flavonoids in Chrysanthemum morifolium could restrain vascular endothelial cell. To some extent, the flavonoids in Chrysanthemum morifolium could prevent and cure the vasculopathy in diabetes, and could provides theory refference.
