目的研究染色体结构维持蛋白4(SMCA)在原发性肝癌组织中的表达及其临床意义。方法收集2009年9月-2010年1月原发性肝癌组织和正常肝脏组织各36份,原发性肝癌患者和健康人血清各36份,通过免疫组化、酶联免疫吸附实验分别检测原发性肝癌组织、正常肝脏组织及血清中SMC4蛋白的表达情况,分析检测结果及其与临床参数之间的关系。结果SMCA在原发性肝癌组织中呈较强染色,其中88.9%(32/36)的肿瘤组织表达阳性,仅11.1%(4/36)的肿瘤组织表达呈阴性,而在正常肝脏组织中未见其染色反应,阴性表达率100%(36/36)。原发性肝癌患者血清中SMCA浓度(255.75±26.59mg/L)远高于健康人(205.89±18.55ng/L,P〈0.05)。统计学分析显示,SMCA的表达与患者肿瘤组织的细胞分化、TNM分期及血管浸润相关(P〈0.05),而与患者的年龄、性别无关(P〉0.05)。结论SMC4在原发性肝癌患者中表达增高,测定其水平有助于原发性肝癌的诊断,并可作为判断肿瘤恶性程度和预后的重要指标。
Objective To study the expression of structural maintenance of chromosomes 4 (SMCA) in primary hepatocellular carcinoma, and to analyze the relationship between the expression with clinicopathological features. Methods From September 2009 to January 2010, samples of primary hepatoeellular carcinoma and normal hepatocellular tissues, serum samples of patients with primary hepatoeellular carcinoma and normal controls were collected from 36 individuals in each group. Immunohistochemical staining was performed to detect the expression of SMC4 of primary hepatocellular carcinoma and normal hepatic tissues, enzyme-linked immunosorbent assay (ELISA) was used to analyze the expression of SMCA of serum samples of patients with primary hepatocellular carcinoma and normal controls. The relationship between results and clinical findings was then analyzed. Results Strong SMCA immunoreactions were detected in primary hepatoeellular carcinoma tissues, with 88. 9% (32/36) positive and 11. 1% (4/36) negative staining, respectively, whereas the immunohistochemical straining was 100% (36/36) negative in normal hepatocellular tissues. The levels of SMCA expression in serum samples from patients with primary hepatocellular carcinoma (255. 75±26. 59ng/L) were significantly higher than those of normal controls (205. 89±18. 55ng/L, P〈0. 05). By statistical analysis, the high levels of SMCA expression in patients with primary hepatocellular carcinoma were found to be related to the cell differentiation, TNM stage and vascular invasion (P〈0. 05), but not to sex and age (P〉 0. 05). Concision High levels of SMCA expression have been detected in primary hepatocellular carcinoma hut not in normal controls, implying that it may be helpful in the diagnosis of the tumor. SMCA can serve as a marker to judge the malignancy and prognosis of patients with primary hepatocellular carcinoma.
