肠道菌群是一个庞大的微生态系统,在维持人体健康方面,具有屏障、免疫、代谢等多种功能,人体多种疾病的发生均伴随有肠道菌群失调症。发酵食品是微生物发酵而制成的一种口味独特、营养价值高的食品,是食品产业的重要支撑。为了充分认识肠道菌群与人体健康关系以及保证发酵产品质量稳定,寻找有效的微生物分离、分析方法尤为重要。本文在简单论述流式细胞技术、PCR-DGGE技术、实时荧光定量PCR技术和焦磷酸测序技术等微生物分离分析方法的基础上,介绍了高效液相色谱技术在该研究领域的应用及其潜力,以期为肠道菌群和发酵食品微生物的分离、分析提供新的思路。
The intestinal flora forms a complex micro-ecological system, which plays an important role in barrier protection, immune response, metabolism and so on;its disorder is closely related to the occurrence of various diseases. Fermented foods that made from microorganism fermentation with unique taste and high nutritional value is a crucial support for food industry. For the purpose of fully understanding the relationship between intestinal flora and hu- man health and ensuring the quality of fermented products, there is a great need for methods of separation and analysis of microorganism. This review describes the application and potential of the HPLC technique in the separation and analysisi of microbes, based on the briefly introduction of some traditional methods, such as flow cytometry, PCRDGGE and FQ-PCR in this field, so as to provide new ideas of the methodology for the analysis, separation and preparation of intestinal microflora and microecology of fermented foods.
