目的应用表面等离子体共振(SPR)芯片检测系统进行淋病奈瑟氏菌的快速检测。方法制备淋病奈瑟氏菌的SPR响应型基因芯片,并对最适的探针自组装时间,探针浓度,以及SPR检测的灵敏度,特异性等指标进行测定;对临床样品进行实际检测并与临床常规方法比较。结果当固定探针浓度为1500aM,自组装时间为4.5h时,构建的芯片具有较好的SPR检测效率,能够准确识别碱基的错配,其检测灵敏度大约为10fM;临床检测结果与常规经典鉴定方法相比结果一致。结论本芯片检测系统具有良好的检测性能,检测方法准确可靠。
Objective Application of SPR-based gene-chip system for rapid detection of Neisseria gonorrhoeae. Methods The SPR-based gene chip ofNeisseria gonorrhoeae was prepared and the optimal luting time and concentration of probes, the sensitivity and the specificity of the chip were verified. Finally, the SPR-based gene chip was utilized to detect clinical samples. Results When fixed by probes for 4.5 h at concentration of 1500nM, the gene chip shows a well detection performance. It can identify accurately the mismatch between bases and its detection sensitivity is 10fmol/L. When compared with the conventional analysis, the detection results of the chip system were essentially the same. Conclusion The detection results proved that this chip system is accurate and reliable. The chip will have a promising prospect in application.
