目的研究芹菜素与柚皮素经大鼠空肠、回肠与结肠黏膜的区段透过特征。方法采用体外Ussing Chamber方法,分别评价50μmol.L-1芹菜素及柚皮素经空肠、回肠和结肠黏膜经时吸收方向(M-S)与分泌方向(S-M)的累计透过率和表观渗透系数(Papp),芹菜素与柚皮素在接收池的样品浓度用HPLC测定。结果芹菜素与柚皮素在结肠的透过性显著高于空肠(P〈0.01)和回肠(P〈0.05)。比较M-S方向和S-M方向的透过性发现,芹菜素空肠与回肠经两个方向的透过性无显著差异,但经结肠透过时,吸收方向的透过性显著高于分泌方向的透过性(P〈0.01);柚皮素在空肠、回肠及结肠黏膜经两个方向的透过性均无显著差异。结论芹菜素与柚皮素在大鼠肠黏膜中的透过性存在区段性差异,结肠部位吸收最好。芹菜素与柚皮素除被动扩散外,可能受到肠黏膜转运体的调控。
OBJECTIVE To investigate the permeabilities of apigenin and naringenin across the jejunal,ileal and colonic membranes in rats.METHODS The cumulative permeation amounts and the permeabilities(Papp) of apigenin and naringenin via rat intestinal membranes in both absorption(M-S) and secretion(S-M) direction at the concentration of 50 μmol·L-1 were evaluated by the method of Ussing Chamber.The concentrations of apigenin and naringenin in the receptor samples were determined by HPLC.RESULTS The permeabilities of apigenin and naringenin across the colonic membrane were significantly higher than those across jejunal(P0.01) or ileal(P0.05) membrane.And the permeability of apigenin in the colonic membrane from the mucosal to serosal direction was significantly greater than that from the mucosal to serosal direction(P0.01).CONCLUSION The permeabilities of apigenin and naringenin across the jejunum,ileum and colon in rats exhibit regional differences,which show the highest in the colon.In addition,the transport of apigenin and naringenin may be regulated by some transporters besides passive diffusion.
