目的检测新疆维吾尔族与汉族哮喘患者外周血CD_4~+T淋巴细胞白介素2(IL-2)、白介素4(IL-4)、干扰素γ(IFN-γ)mRNA的表达水平,比较其表达差异在疾病中的作用。方法选取2014年12月—2015年5月在新疆医科大学第一附属医院和新疆维吾尔自治区人民医院确诊的哮喘急性发作期患者150例为哮喘组(包括维吾尔族30例、汉族120例),及同期在新疆医科大学第一附属医院进行体检的健康人群101例为对照组(包括维吾尔族35例、汉族66例)。应用实时荧光定量反转录聚合酶链反应(RT-PCR)技术检测所有受试者IL-2、IL-4、IFN-γmRNA的表达水平。结果选择内参基因GAPDH进行校正。哮喘组与对照组受试者IL-2、IL-4 mRNA的表达水平间差异有统计学意义(P〈0.05);而IFN-γmRNA的表达水平间差异无统计学意义(P〉0.05)。维吾尔族与汉族哮喘患者IL-2 mRNA的表达水平间差异无统计学意义(P〉0.05);而IL-4、IFN-γmRNA的表达水平间差异有统计学意义(P〈0.05)。汉族哮喘亚组与汉族对照亚组受试者IL-2、IL-4 mRNA的表达水平间差异有统计学意义(P〈0.05);而IFN-γmRNA的表达水平间差异无统计学意义(P〉0.05)。维吾尔族哮喘亚组与维吾尔族对照亚组受试者IL-2、IL-4 mRNA的表达水平间差异有统计学意义(P〈0.05);而IFN-γmRNA的表达水平间差异无统计学意义(P〉0.05)。结论哮喘患者IL-2基因表达高于健康人群,IL-4基因表达低于健康人群;汉族哮喘患者IL-4、IFN-γ基因表达均低于维吾尔族哮喘患者。提示IL-2、IL-4、IFN-γ基因在哮喘发病过程中发挥重要作用。
Objective To examine the expression of IL-2,IL-4 and IFN-γ mRNA in CD_4~+T lymphocytes in the peripheral blood of Uygur and Han asthma patients in Xinjiang. Methods We enrolled 150 patients( 30 Uygur patients and 120 Han patients) who were diagnosed with asthma at acute stage in the First Affiliated Hospital of Xinjiang Medical University and Xinjiang People's Hospital from December 2014 to May 2015 as asthma group. We also enrolled 101 healthy people( 35 Uygur people and 66 Han people) who received physical examination in the the First Affiliated Hospital of Xinjiang Medical University in the same period. Quantitative real- time RT- PCR was applied to detect the expression levels of IL-2,IL-4 and IFN-γ mRNA in all subjects. Results Reference gene GAPDH was selected to make adjustment. Asthma group and control group were significantly different in the expression of IL-2 and IL-4 mRNA( P〈0. 05),but were not significantly different in the expression of IFN-γmRNA( P〈0. 05). Uygur and Han asthma patients were not significantly different in the expression of IL-2 mRNA( P〈0. 05),but were significantly different in the expression of IL-4 and IFN-γ mRNA( P〈0. 05). Han asthma subgroup and Han control subgroup were significantly different in the expression of IL-2 and IL-4 mRNA( P〈0. 05),but were not significantly different in the expression of IFN-γ mRNA( P〈0. 05). Uygur asthma subgroup and Uygur control subgroup were significantly different in the expression of IL-2 and IL-4 mRNA( P〈0. 05),but were not significantly different in the expression of IFN-γ mRNA( P〈0. 05). Conclusion The expression of IL-2 gene of asthma patients is higher than that of healthy people,and the expression of IL-4 gene is lower than that of healthy people. Han asthma patients are lower than Uygur asthma patients in IL-4 and IFN-γ gene expression. IL-2,IL-4 and IFN-γ genes play an important role in the pathogenic process of asthma.
