【目的】研究claudin11在胃癌中的表达及临床价值,预测claudin11参与肿瘤发生发展的机制。【方法】从癌症基因组图谱(the cancer genome atlas,TCGA)和人类肿瘤相关的基因表达汇编(gene expression omnibu,GEO)数据库收集胃癌数据集,下载claudin11基因表达谱及临床信息。分析claudin11表达和临床病理学参数的相关性及对预后的影响。采用基因集富集分析(gene set enrichment analysis,GSEA)方法预测claudin11的调控基因。【结果】在TCGA数据集中claudin11表达水平与年龄(P=0.031)、T分期(P=0.007)、有无淋巴结转移(P=0.039)、TNM分期(P=0.03)及分化程度(P〈0.001)相关。Claudin11高表达患者总生存期明显短于低表达患者(P〈0.001)。Claudin11高表达样本富集了细胞黏附、紧密连接、细胞外基质、骨架蛋白调控等相关等基因集。【结论】Claudin11在胃癌中是一种不良预后因素,可以作为判断预后的标志物。
【Objective】To claudin11 expression in gastric cancer and its clinical value, and predict the role of claudin11 participating in the genesis and development of tumors.【Methods】The datasets of gastric cancer were collected from The Cancer Genome Atlas(TCGA) and Gene Expression Omnibus(GEO) database. The gene expression atlas of claudin11 and clinical information were downloaded. The correlation between claudin11 expression and clinical pathological characteristics and the effect on prognosis were analyzed.The regulatory gene of claudin11 was predicted by Gene Set Enrichment Analysis(GSEA).【Results】In the TCGA, the expression of claudin11 was correlated with age(P=0.031), T stage(P=0.007), N stage(P=0.039), TNM stage(P=0.03) and differentiation degree(P0.001). High expression of claudin11 indicated poor prognosis(P0.001). The related gene atlases on cell adhesion, tight junction, extracellular matrix and cytoskeleton regulation were rich in the high expression samples of claudin11.【Conclusion】The expression of claudin11 indicates poor prognosis of gastric cancer and can be used as a potential prognostic marker.
