目的观察不同浓度柔红霉素(DNR)对Jurkat细胞生物学特性的影响。方法采用不同浓度的DNR作用Jurkat细胞,观察细胞的生长状况,在不同的时间点收获细胞,检测DNR作用前后Jurkat细胞生物学特性的改变。结果当0.5μmol/LDNR作用12h或18h,可以保证低死亡率的同时,细胞发生凋亡的比率最高;DNR抑制Jurkat细胞bcl-2、PCNA蛋白的表达,使更多细胞受阻于G0/G1期。结论在合适的DNR浓度及作用时间条件下,可能通过抑制Jurkat细胞bcl-2、PCNA蛋白的表达达到诱导凋亡、抑制增殖的作用。
Objective To observe the various concentration daunorubicin (DNR)'s influence on biological characteristics of Jurkat cell lines. Methods To treat the Jurkat cells by various concentration DNR,observe their growth status and ingather the cells in different time points. Then the change of biological characteristics were examined. Results When treated with 0.5/Lmol/L DNR for 12h to 18h,the percentage of apoptotic Jurkat cells was the highest and the death rate was the lowest compared with other time points groups. DNR could inhibit bcl-2 and PCNA protein expression and arrest more cells in G0/G1 phase. Conclusion Treated with DNR in proper concentration and for proper action time, Jurkat cells can be efficiently induced to apoptosis and their proliferation can be inhibited, which may be resulted in inhibiting the expression of the bcl-2 and PCNA protein.
