为实现高效液相色谱法高效定量分析李果实中的ATP、ADP和AMP,通过对流动相的组成、p H值、流速、柱温、检测器波长的比较及其他条件的探讨,确定了高效液相色谱法测定3种能量物质的最佳色谱条件。结果表明,采用p H6.25的0.05 mol/L的磷酸二氢铵溶液(含3.0%甲醇)作为流动相,流速1.0 m L/min,柱温35℃,3种能量物质在30 min内有效分离。经方法学验证,线性范围为1~15 mg/L,回收率在52.18%~72.86%;RSD≤5.56%。该色谱条件可为定量测定李果实中ATP、ADP和AMP含量提供技术支持。
In order to establish a HPLC method for the determination of ATP,ADP and AMP,mobile phase composition,mobile phase p H,flow rate,column temperature and detection wavelength were optimized.The mobile phase was 0.05 mol/L ammonium dihydrogen phosphate solution(containing 3.0% methanol,p H6.25).The flow rate was 1.0 m L/min and the column temperature was 35 ℃.3 adenosine phosphates were completely separated within 30 min.The linear range of the method was 1~15 mg/L.The average recovery rate was 52.18%~72.86% and RSD≤5.56%.The optimization could provide technical support for the determination of ATP,ADP and AMP content in plum fruit.
