目的:建立一种牙周炎合并血管钙化复合动物模型。方法:20只成年Wistar雄性大鼠随机分为血管钙化+慢性牙周炎组(VDN+cP)和正常对照组(C)(n=10);VDN+CP组采用丝线局部结扎、牙周混合致病菌感染合并维生素D,肌注和尼古丁灌胃的方法建立VDN+CP模型,建模第8周检测各牙周临床指标,然后处死各大鼠,取心脏称重并测量心重指数;取胸主动脉制备石蜡切片,HE染色观察血管改变;取含上颌磨牙的上颌骨,CBCT扫描观察牙槽骨吸收情况。结果:VDN+CP组牙周炎症明显,牙周探诊深度(probingdepth,PD)、龈沟出血指数(sulcular bleeding index,SBI)、牙槽骨丧失值以及心重指数均明显高于C组(P〈0.05);血管病理学观察显示:VDN+CP组胸主动脉形成血管钙化病变,C组未见明显异常改变。结论:通过维生素职肌注加尼古丁灌胃联合丝线结扎和细菌接种法初步成功建立了慢性牙周炎合并血管钙化动物模型。
AIM: To establish an animal model of vascular calcification with chronic periodontitis (VDN + CP) in Wistar rats. METHODS :20 male Wistar rats were divided into 2 groups randomly( n = 10). The rats in VDN + CP group were treated by intramuscular injection of vitamin D3 , intragastric administration of nicotine, silk ligation and mixed bacteria infection. After 8 weeks of treatment probing depth (PD) and sulcular bleeding index (SBI) were measured. Then animals were sacrificed, heart weight/body weight (HW/BW) was measured. Maxillae including mo- lars were sectioned and scanned by Cone beam CT. Vascular tissue was examined by HE staining. RESULTS: PD and SBI of VDN + CP group were higher than those of the control group ( P 〈 0.05 ). The alveolar bone resorption of VDN + CP group was more than that of control group ( P 〈 0.05 ). Vascular calcification lesions were found in aorta pectora- lis in the rats of of VDN + CP group. HW/BW of VDN + CP group was higher than that of the control group ( P 〈 0.05 ). CONCLUSION : The animal model of chronic periodontitis accompany with vascular calcification can be successfully established by administration of vitamin D3 and nicotine, silk ligation and mixed oral pathogenic bacteria infection.
