A new ruthenium(II) polypyridyl complex of [Ru(bpy)2(hedppc)](ClO4)2 {in which bpy= 2,2′-biyridine, hedppc=2-hydroxyethyl dipyrido[3,2-a:2′,3′-c]phenazine-ll-carboxylate} was synthesized and characterize.d by elemental analysis, ^1H NMR and MALDI-TOF MS. The binding of the complex to calf thymus (ct) DNA was studied with UV-visible absorption and luminescence spectrophotometric titrations, DNA competitive binding with ethidium bromide, viscosity measurements and thermal denaturation experiments. The results indicated that the complex was bound to ct-DNA through an intercalative mode with an intrinsic binding constant of (6.99± 1.34)× 10^6 L/mol in buffered 50 mmol/L NaCl comparable to parent [Ru(bpy)2(dppz)]^2+. The complex is permanently non-emissive in the absence and presence of the DNA, and exhibits intriguing solvatochromic properties different from parent [Ru(bpy)2(dppz)]^2+.
