目的探讨电针治疗阿尔茨海默病的可能作用机制。方法将快速老化小鼠P8亚系(SAMP8)36只随机分为模型组、电针组和药物组,抗快速老化小鼠R1亚系(SAMR1)12只为正常组。电针组给予电针百会穴、印堂穴20min后,点刺水沟穴;药物组给予盐酸多奈哌齐按0.92mg·kg-1剂量灌胃;正常组和模型组给予电针组相同程度和时间的束缚。治疗15d后,用Morris水迷宫测定小鼠学习记忆能力,免疫组化技术检测小鼠大脑皮层B淀粉样蛋白(Ap)及β位淀粉样前体蛋白裂解酶(BACE-1)含量,实时荧光定量PCR法测定BACE-1mRNA的表达。结果与模型组比较,药物组与电针组逃避潜伏期均显著下降,而目标象限游泳时间均显著增加(P〈0.05),电针组BACE-1、BACE-1mRNA表达显著减少(P〈0.05);电针组游泳总路程、游泳速度明显下降,与药物组比较差异有统计学意义(P〈0.05)。结论电针可能通过抑制SAMP8小鼠大脑皮层BACE-1表达,降低AB水平,达到改善其学习记忆障碍的作用。
Objective To discuss the potential effect mechanism on Alzheimer~s disease with dec- troacupuncture. Methods Thirty - six mice of senescence - accelerated mice prone 8 ( SAMP8 ) were ran- domized into a model group, an electroacupuncture group and a medicine group. Twelve mice of senescence accelerated mouse/resistance 1 ( SAMR1 ) were in a normal group. In the electroacpuncture group, electroacu- puncture was applied to Baihui (GV20) and Yintang ( EX - HN3 ) for 20min. Afterwards, the pricking tech- nique was used at Shuigou (GV26). In the medicine group, the gastric infusion with Donepezil HC1, at 0. 92mg/kg was applied. In the normal group and the model group ,the same degree and time restriction were a- dopted as the electroacupuncture group. After treatment for 15 days, Morriz water maze test was used to deter- mine the learning and memory ability of mice. IHC technology was used to determine the contents of 13 amy- loid protein(Aβ)and β amyloid precursor protein cleaving enzyme (BACE -1 ). The real -time fluorescent quantitative PCR was applied to determine the expression of BACE - 1 mRNA. Results Compared with the model group, the escaping latent period was reduced significantly in the medicine group and the electroacu- puncture group, and the target quadrant swimming time was increased significantly ( P 〈 0.05 ) ; the expres- sions of BACE - 1 and BACE - 1 mRNA were reduced significantly in the electroacupuncture group ( P 〈 0. 05 ). The total distance and velocity of swimming were reduced apparently in the electroacupuncture, indica- ting the significant difference as compared with the medicine group (P 〈 0.05 ). Conclusion Electroacu- puncture has the improvements in learning and memory disturbance probably through inhibiting BACE - 1 ex- pression and reducing Aβ level of cerebral cortex in SAMP8 mice.
