目的研究三参方对慢性萎缩性胃炎(CAG)大鼠胃组织表皮生长因子(EGF)表达及血清、胃黏膜一氧化氮(NO)含量和一氧化氮合酶(NOS)活性的影响,以探讨其治疗CAG的作用机制。方法将60只雄性Wistar大鼠按体质量分层随机分为6组,即正常对照组、模型组、维酶素组、三九胃泰组及三参方高(3.67 g/mL)、低(1.83 g/mL)剂量组,每组各10只。正常对照组予蒸馏水灌胃,自由饮食、饮水;其余各组采用复合因素造模法复制大鼠CAG模型,连续8周。造模成功后,三参方高、低剂量及维酶素组、三九胃泰组分别予相应药液10 mL/(kg.d)灌胃,正常对照组及模型组予等容积蒸馏水灌胃,连续35 d。观察各组大鼠胃组织EGF表达及大鼠血清、胃黏膜NO含量和NOS活性、胃黏膜组织形态变化情况。结果模型组大鼠胃黏膜上皮细胞可见不同程度的变性、坏死、脱落,腺体有不同程度的萎缩,间质内有大量炎性粒细胞浸润。各药物组大鼠胃黏膜病变均有一定程度改善,其中三参方高剂量组大鼠胃黏膜腺体大小形态较一致,基本接近正常对照组。模型组大鼠胃黏膜EGF表达的OD值较正常对照组明显增强(P〈0.01),血清、胃黏膜NO含量和NOS活性明显升高(P〈0.01)。维酶素组、三九胃泰组及三参方高、低剂量组OD值表达较模型组明显减弱(P〈0.05,P〈0.01),血清、胃黏膜NO含量和NOS活性降低(P〈0.05,P〈0.01),其中三参方高剂量组OD值表达与其他各治疗组比较均明显减弱(P〈0.01),胃黏膜NO含量低于维酶素组(P〈0.05),血清NO含量低于三九胃泰组(P〈0.05);三参方低剂量组及维酶素组OD值表达均较三九胃泰组弱(P〈0.05),三参方高、低剂量组和三九胃泰组大鼠血清NO含量低于维酶素组(P〈0.01,P〈0.05)。结论三参方可通过抑制胃组织EGF的过度表达,降低NOS活性,抑制血清、胃黏膜NO的过度释放,促?
Objective To explore the effect of Sanshenfang on epidermal growth factor expression of the gastric tissue and content of carbon monoxide and nitric oxide synthase(NOS) activity in serum and gastric mucosa of rat model with chronic atrophic gastritis.Methods 60 male Wistar rats,stratified by body mass were randomly divided into six groups,the normal control group,model group,Vitacoenzyme group,Sanjiuweitai group and Sanshenfang high-(3.67 g/mL),low-dose(1.83 g/mL) group.Rats in normal control group were given distilled water,free food,drinking water.Remaining groups were copy the model of rat CAG by composite factors modeling method for 8 weeks.Sanshenfang high-,low-dose,Vitacoenzyme and Sanjiuweitai were given in different groups after the modeling for 35 days.EGF expression of the rat's gastric tissue,NO content and NOS activity in serum and mucosa and gastric mucosa morphological changes were observed.Results Rat gastric mucosal lesions have a certain degree improvement in each drug group,which size and shape of rat gastric mucosa glands in Sanshenfang high-dose group is more consistent with the normal control group.OD values of rat gastric mucosa VEGF expression in the model group were significantly enhanced in comparison with normal group(P0.01).NO content and NOS activity in serum and mucosa was significantly higher(P0.01).OD values of rat gastric mucosa VEGF expression in,Sanjiuweitai group and Vitacoenzyme group were significantly weakened as compared with that in model group(P0.05,P0.01),NO content and NOS activity in serum and mucosa was decreased as compared with that in model group(P0.05,P0.01).OD value of the expression in Sanshenfang high-dose group were significantly weakened as compared with that in the other treatment group(P0.01),NO content in mucosa was lower than that in Vitacoenzyme group(P0.05),NO content in serum in was lower than that in Sanjiuweitai group(P0.05).NO content in serum Sanshenfang high-,low-dose group and Sanjiuweitai group was lower
