目的:研究两色金鸡菊提取物对实验性糖尿病模型小鼠血糖血脂以及胰腺组织结构的影响。方法:采用60%乙醇提取、乙酸乙酯萃取的方法得到两色金鸡菊提取物;采用高糖高脂饲料喂养加STZ的方法建立实验性糖尿病小鼠模型;将建立的模型小鼠按血糖分为模型组、二甲双胍组(0.16g/kg)、两色金鸡菊提取物8.0g/kg、4.0g/kg、2.0g/kg剂量组,连续灌胃30d。每周测量各组小鼠空腹血糖(FBG),干预30后检测各组小鼠血清中C肽、TC、TG、LDL-C、HDL-C含量,计算胰岛素抵抗指数(IRI)和胰岛素分泌指数,HE染色观察各组小鼠胰腺组织结构的变化。结果:两色金鸡菊提取物8.0g/kg剂量组可明显降低糖尿病小鼠的FBG、TC、TG、IRI,增加血清中C肽的含量,明显改善胰岛β细胞功能;病理结果显示,两色金鸡菊提取物对损伤的胰岛具有一定的恢复作用。结论:两色金鸡菊提取物具有降低实验性糖尿病小鼠的血糖、改善血脂的作用,其作用机制可能与恢复胰岛β细胞功能,促进胰岛素分泌有关。
Objective: To study the effect of the ethyl acetate extract from Coreopsis tinctoria nutt. on fasting blood levels of glucose,lipids and pancreatic pathology of the diabetic mice. Methods: Diabetic mice model was replicated by feeding high-fat and high-sugar diet combined with streptozotocin( STZ) injection. The diabetic mice were randomly divided into five groups,untreated control,the high,middle,low dosage group of the ethyl acetate extract( 8. 0 g /kg,4. 0 g /kg,2. 0 g /kg),metformin group( 0. 16 g /kg). Fasting blood glucose and body weight were measured every week. After 30 days,the levels of fasting blood glucose,triglycerides( TG),total cholesterol( TC),high-densitylipoprotein cholesterol( HDL-C),low-densitylipoprotein cholesterol( LDL-C),and C peptide were examined. Calculate the insulin resistance index( IRI) and insulin secretion index and observed the pancreatic pathology after treating with ethyl acetate extract for 30 days. Result: Compared with model group,he high-dose of the ethyl acetate extract treatment significantly decreased the levels of FBG,TG,TC,IRI,while the content of C peptide and Insulin secretion index were increased. The damaged pancreatic pathology also be repaired. Conclusion: The ethyl acetate extract from Coreopsis tinctoria Nutt. could reduce the fasting blood levels and lipids,which might be related to improve islet β cell function and promote the secretion of insulin.
