两色金鸡菊提取物对大鼠肾小球系膜细胞增殖及纤维化因子表达的影响

目的建立高脂高糖与低剂量链脲佐菌素(STZ)诱导的SD大鼠的早期糖尿病肾病(DN)模型,模拟早期DN的病理改变及肾功能改变。方法将SPF级正常雄性SD大鼠30只按照体重随机分为正常组、模型组。正常组大鼠给予普通饲料喂养,模型组大鼠高脂饲料喂养8周后,腹腔注射STZ 35 mg/kg,形成糖尿病模型后,继续高脂高糖饲料喂养4周后,观察两组大鼠肾脏病理改变、生化及肾功能各项指标的改变情况。结果模型组大鼠体重显著低于正常组(P〈0.01),而随机血糖值显著高于正常组(P〈0.01),且达到糖尿病成模标准。模型组大鼠血清胰岛素水平显著低于正常组,相对肾重、24 h尿微量白蛋白、α1-微球蛋白、β2-巨球蛋白值显著高于正常组,差异有高度统计学意义(P〈0.01)。模型组病理特征表现为肾小球基底膜增厚,系膜增宽,肾小球及肾小管糖原物质、胶原纤维聚集等。结论此造模方法可模拟早期DN的基本特征。

英文摘要:

Objective To establish the early diabetic nephropathy(DN) rat model induced by high-fat and high-glucose and low dose of Streptozocin(STZ), so as to simulate the pathologic and renal function changes of early DN.Methods Thirty SPF level of normal male SD rats were divided into normal group and model group by weight.Normal group was fed with normal diet, model group was fed with high-fat and high-glucose diet for 8 weeks continuously,then 35 mg/kg of STZ was injected intraperitoneally to establish diabetic rats model, after that, high-fat and high-glucose diet was fed for 4 weeks continuously.Renal pathological changes, biochemical and renal function indexes of two groups were observed.Results The body weight of rats in the model group was significantly lower than that of normal group(P 〈0.01), while the random blood glucose level was higher than that of normal group(P 〈0.01), which had reached to standard of diabetes mellitus.The level of serum insulin in the model group was lower than that of normal group, the values of relative kidney weight, 24 h microalbuminuria, α1-micr oglobulin, β2-macroglobulin were higher than those of normal group, the differences were highly statistically significant(P 〈0.01).The pathological characteristics of model group presented the thickening of glomerular basement membrane, mesangial broadening, accumulation of glycogen and collagen fiber in both glomerulus and kidney tubules.Conclusion This molding method can imitate the basic characteristics of early DN.
