目的:探讨仪ανβ6整合素介导的细胞与细胞外基质的黏附,及其对氟尿嘧啶诱导的结肠癌细胞凋亡的影响。方法:采用流式细胞仪测定结肠癌细胞株HT-29和WiDr细胞ανβ6整合素的表达,并应用酶联免疫吸附实验(ELISA)及丫啶橙(AO)-溴化乙啶(EB)双荧光染色方法检测细胞凋亡。结果:HT-29细胞、WiDr细胞均表达ανβ6整合素,表达率分别为80.82%及82.96%。氟尿嘧啶诱导结肠癌HT-29和WiDr细胞凋亡。经氟尿嘧啶(20mg/L)作用48h后,酶联免疫吸附实验结果显示,生长于ανβ6整合素配体-纤黏连蛋白(FN)上的HT-29细胞、WiDr细胞的富集系数(EF)分别为1.11±0.04、1.09±0.02,明显低于生长于非整合素配体-多聚赖氨酸(polylisin)上的细胞分别为3.68±0.03、3.72±0.02,P<0.01;用ανβ6整合素的特异性单抗封闭后再生长于FN上时,HT-29、WiDr细胞的EF分别为2.12±0.04、2.14±0.03,明显高于直接生长于FN上(P<0.01)。AO—EB双荧光染色法结果显示,生长于FN上的HT-29和WiDr细胞凋亡率分别为(5.6±1.1)%、(5.3±0.7)%,明显低于生长于polylisin上的细胞的凋亡率(37.0±1.4)%、(38.5±0.9)%,P<0.01;用ανβ6整合素的特异性单抗封闭后再生长于FN上,细胞凋亡率分别为(19.5±1.2)%、(20.0±0.7)%,又有明显提高(P<0.01)。结论:结肠癌细胞株HT-29和WiDr细胞表达ανβ6整合素,ανβ6整合素介导的细胞与FN的黏附抑制氟尿嘧啶诱导的结肠癌细胞凋亡。提示细胞黏附可能通过抑制细胞凋亡而促使细胞产生耐药性。
AIM : To investigate the effects of ctv136 integrin - mediated cell adhesion on 5 - fluorouracil (5 - FU) induced apoptosis in colon carcinoma cell lines. METHODS: The expression of the ανβ6 integrin in colon carcinoma cell lines HT - 29 and WiDr cells was analyzed by flow cytometry. The apoptosis induced by 5 - FU and the effects of ανβ6 integrin - mediated cell adhesion on 5 - FU induced cell apoptosis were measured by enzyme - linked immunosorbent assay (ELISA) method and acridine orange -ethidium bromide (AO -EB) double fluorescent dye staining. RESULTS:Both the colon carcinoma cell lines HT - 29 and WiDr cells expressed the ανβ6 integrin. The percentages of HT - 29 and WiDr cells expression were 80. 82% and 82.96%. 5 - FU induced the apoptosis of colon carcinoma cell lines HT -29 and WiDr. The result of ELISA method displayed that enrichment factor (EF) of HT -29 and WiDr cells planted on fibronectin (FN) -ligand of ανβ6 integrin was lower significantly than the EF of HT -29 and WiDr cells planted on non -integrin ligand polylisin ( 1.11 ± 0. 04 vs 3.68 ± 0. 03, 1.09 ± 0. 02 vs 3.72 ± 0.02, P 〈 0. 01 ) after cultured in medium containing 20 mg/L 5 - FU for 48 h. When HT - 29 and WiDr cells preineubated with ανβ6 integrin blocking antibody were planted on FN again, the EF of HT - 29 and WiDr cells was higher significantly than those directly planted on FN without being blocked by ανβ6 integrin antibody (2. 12 ± 0. 04 vs 1.11±0. 04, 2. 14 ± 0. 03 vs 1.09 ± 0.02, P 〈 0. 01 ).
