为了给苔藓结皮野外恢复的大规模接种提供丰富种源,本文利用0.1%NaClO(10、15、20、30、45、60、120s)和0.1%HgCl2(10、15、20、30、45、60、120s)两种消毒液,采用Knop、MS和Hoagland3种培养基,设置5.5、6、6.5、7、7.5、8、8.5七个水平的pH值,通过单因素试验设计方法,测定长尖对齿藓[Didymodon ditrichoides(Broth.)]茎叶体的成活率、原丝体长度和分枝数等指标,探讨影响长尖对齿藓原丝体扩繁的关键因子。结果表明:(1)消毒方式、培养基及pH均对长尖对齿藓茎叶体的成活率、原丝体长度和分枝数有显著影响(P〈0.05),影响大小依次均为消毒方式〉培养基〉pH;(2)最适合的消毒方式为:0.1%NaClO消毒15~20s;(3)最适合长尖对齿藓原丝体生长的培养基是Knop和Hoagland培养基;(4)最适合长尖对齿藓原丝体生长的pH是7.5。从而得出快速培育长尖对齿藓原丝体的最佳组合为:0.1%NaClO消毒15~20s+Hoagland/Knop+pH=7.5。本文的研究结果可以缩短长尖对齿藓生长周期以进行快速繁殖,为苔藓结皮的快速培育以及工程化应用提供一定的借鉴。
In order to provide a rich source of mass vaccination for the field restoration of moss crust, we used two kinds of disinfection method, 0.1% NaClO(10, 15, 20, 30, 45, 60, 120 s) and 0.1% HgCl2(10, 15, 20, 30, 45, 60, 120 s) , with Knop, MS and Hoagland as culture media, and set the pH values(5.5, 6, 6.5, 7, 7.5, 8, 8.5) , through the single factor experiment design method, we determined the survival rate of the cormus of [ Didymodon ditrichoides (Broth.)], protonema length and number of branches, thereby exploring key factors that affect the propagation of the protonema of 1). ditrichoides. The results are as follows : ( 1 ) The disinfection method, culture medium and pH value have significant influence upon the survival rate of the corrnus of D. ditrichoides, length of protonema and number of branches ( P 〈 0.05 ) , in a descending order of disinfection method, culture medium, and pH value ; (2) The most suitable disinfection method is 0.1% NaClO with a duration of 15 -20 s; (3)The most suitable culture medium for the growth of the protonema of D. ditrichoides is Knop and Hoagland ; (4) The most suitable pH value for the growth of protonema of D. ditrichoides is 7.5. Therefore, the best combination of factors for rapid cultivation of the protonema of D. ditrichoides is 0. 1% NaClO disinfection 15 - 20 s + Hoagland/Knop + pH = 7.5. Our experiment can be determined to shorten the growth cycle of D. ditrichoides and realize rapid propagation, thereby providing experience for the rapid culture and engineering application of moss crust.
