目的总结细胞损伤模型在化合物抗脑缺血活性筛选中的应用,旨在对药物的抗脑缺血研究提供新思路。方法参考国内外相关文献29篇,阐述急性脑缺血相关的基本理论以及目前常用的三种模拟体内病理进程的脑缺血体外细胞损伤模型的方法,并结合此三种细胞损伤模型,对细胞损伤模型如何用于新化合物的活性筛选进行了探讨。结果考察的三种体外模型涵盖了脑缺血时级联反应的三步重要进程,其中缺氧缺糖后复灌模型是最初损伤进程,而谷氨酸与H2O2模型是导致脑缺血损伤最严重的两步进程,模型符合脑缺血后病理损伤理论,具有一定的理论依据。结论可以利用三种模型初步判断化合物作用机制。
Objective To sum up the application of cell injury models in compounds anti-cerebral ischemia activity screening in order to provide a new idea for the research of anti-cerebral ischemia in medicine.Methods The basic theories which related to acute cerebral ischemia and three cerebral ischemia in vitro cell injure models that used widely to imitate in vivo pathological mechanism progress were expounded.The way that the cell injure models used in new compounds' activity screening was discussed combined with these three cell injure models.Results Three cell injury in vitro models were used,which were now most widely accepted for they could represent three main pathophysiological mechanisms of ischemic cascade.OGD/R induced injury model was the initial injury process,excitotoxicity and oxidative stress had the most serious affects on the final ischemic damage.All models accorded with the pathophysiological injury theory for ischemic stroke and had certain representativeness,and the screeing results were thus comprehensive and might be reliable.Conclusions This method can be used to judge the impact mechanism of compound initially.
