目的 从缺血性脑卒中后大鼠边缘系统碱性成纤维生长因子( bFGF)的时相变化,分析和探讨bFGF在脑卒中后精神症状发生发展中的作用及益肾调气中药的干预环节. 方法 选择雄性Wistar大鼠,经Open-Field行为学评分后,随机分为正常组、假手术组、多发脑梗( MCI )组、中药组、西药组. 正常组常规饲养,假手术组经颈外动脉推注0 .3 mL生理盐水. 模型及治疗组均采用同种系微栓子体外注入法,由颈外动脉推注同等剂量的同种异体血栓栓子;中药组及西药组在模型制备后,分别予益肾调气中药、尼莫地平干预给药4周. 模型复制成功后,于第1、2、4周观察边缘系统bFGF的变化及中药干预效果. 结果 bFGF免疫组化染色结果显示, MCI组第1周bFGF表达明显升高,之后逐渐下降,表明7 d为其表达高峰期. 中药组bFGF表达明显高于正常组及MCI组,提示中药可上调bFGF表达. 结论 脑卒中后大鼠存在边缘系统bFGF的改变,益肾调气中药能加速血管的修复和再生,改善神经元存活微环境而起到恢复受损功能,从而达到阻断精神症状发生的作用.
Objective To investigate the time-course expressions of the basic fibroblast growth factor ( bFGF) in limbic system of cerebral ischemic rats and explore the effects of Yishen Tiaoqi ( kidney-tonifying qi-regulating) Formula.Methods Male Wistar rats were randomly divided into normal group , sham-operation group, MCI group, treatment group and control group according to the scores of Open-Field test. The normal group received no intervention. The sham-operation group received sham operation, and intragastric administration of distilled water.Homogenous microthrombus were injected into internal carotid artery through the external carotid artery of those rats in other groups to duplicate MCI model first.The treatment group and control group received intragastric administration of Yishen Tiaoqi Formula (6.2g/kg) and Nimodipine (9.45mg/kg) respectively.The changes of limbic bFGF and intervention efficacy of Chinese medicinal were observed by using immunohistochemistry technique in the 1st Week,2nd Week and 4th Week.Results Immunohistochemistry test showed that positive cells of bFGF increased significantly in MCI group.The expression of bFGF was significantly higher in the first week than in other time points in MCI group, reached peak at day 7 and then decreased gradually.The positive cells of bFGF increased significantly in Chinese medicinal group compared with MCI group and normal group in the 2nd Week and 4th Week.Conclusion There are changes of limbic bFGF in rats after cerebrovascular disease, and Yishen Tiaoqi Formula can block mental disorder after stroke with improved vascular repair and regeneration.
