目的从脑卒中后大鼠精神症状组群的改变探讨脑卒中后精神症状的演变趋势及益肾调气方药——颐脑解郁方对其早期干预作用。方法选择雄性Wistar大鼠,经Open-Field行为学评分后,随机分为正常组、假手术组、多发脑梗(MCI)组、中药组、西药组、卒中后抑郁(PSD)组及血管性痴呆(VD)组。正常组常规饲养。假手术组经颈外动脉推注0.3mL生理盐水,并灌胃蒸馏水。其余各组均采用同种系微栓子体外注入法,由颈外动脉推注同等剂量的同种异体血栓栓子;中药组及西药组在模型制备后,予颐脑解郁方、尼莫地平干预给药4周;PSD组术后7d予21d慢性不可预知的温和应激(CUMS)加孤养复制PSD模型;VD组术后28d经水迷宫筛选后进入实验。模型复制成功后,于第4周进行表征观察包括动态观察大鼠毛发、眼部变化,平均进食量及大鼠体重;同时进行行为学测试:蔗糖水消耗实验、Open—Field行为测定、穿梭箱主动回避反应实验。结果表征观察发现3个模型组大鼠平均进食量下降,尤以PSD组及‘VD组下降显著,与体重变化趋势相一致。行为学测定发现3个模型组大鼠存在兴趣及记忆能力明显下降;蔗糖水消耗量、Open-Field实验提示PSD组以兴趣下降为主;穿梭箱主动回避反应实验结果提示VD组记忆及学习能力下降更为明显。而中药能减缓表征、学习和兴趣能力的下降趋势。结论脑血管病后大鼠存在精神症状组群的改变,并有向PSD及VD演变趋势。颐脑解郁方能减慢此过程的进展。
Objective To probe into the development trends of post-stroke mental symptoms based on the changes of post-stroke mental symptom group in rats and the interventional effect of Yi Nao Jie Yu Fang with the actions of replenishing kidney and regulating qi. Methods Male Wistar rats, after open-field ethology scoring, were randomly divided into the normal group, sham-operation group, multiple cerebral infarction (MCI) group, Chinese medicinal group, Western medicinal group, post-stroke depression (PSD) group and vascular dementia (VD) group. The normal group was fed normally, sham-operation group was injected normal saline solution (0. 3 mL) through the external carotid artery and given distilled water intragastrically, and other groups were injected homogenous heterophile thromboembolus through the external carotid artery by using the injection of homogenous microembolus in vitro. The Chinese medicinal group was given Yi Nao Jie Yu Fang and Western medicinal group was given nimodipine all for 4 weeks after modeling. After the operation for 7 days, the PSD group was given chronic unpredictable mild stress (CUMS) and isolation rearing for 21 days for duplicating PSD model. After the operation for 28 days, the VD group was selected by Morris water maze and then put into the test. On the 4th week after successfully modeling, the characterization observation was carried out in rats including the changes of hair and eyes, average food intake and body weight. At the same time the ethological detection was conducted including consumption test of sucrose water, open-field test, and test of shuttle box. Results The characterization observation showed that the average food intake decreased in the MCI group, PSD group and VD group, especially in the latter two groups, and the change trend of body weight was similar.
