目的:研究宁夏地区栽培甘草(Glycyrrhiza uralensis Fisch.)中获得的甘草总黄酮(licorice flavonoids,LF)的抗应激抑郁作用的机制。方法:将40只雄性SD大鼠分为正常组对照组、模型组、甘草总黄酮组(100、300 mg/kg)和氟西汀对照组(10 mg/kg);建立慢性不可预见性应激抑郁模型(chronic unpredicted stress model of depression,CUS)。采用强迫游泳实验(forced swimming test,FST)、悬尾实验(tail suspension test,TST)和旷场实验(open field test,OFT)评价甘草总黄酮的抗应激抑郁行为作用;Western蛋白印迹法检测海马组织中Bcl-xl和Caspase-3蛋白表达水平的变化。结果:甘草总黄酮能够明显改善CUS大鼠的应激抑郁行为;海马中Bcl-xl蛋白表达明显增加,Caspase-3蛋白表达明显降低。结论:甘草总黄酮抗大鼠慢性应激抑郁行为的作用可能涉及神经细胞凋亡的保护机制。
Objective: To investigate the antidepressant effect and related mechanism of the total flavonoids extract parts( licorice flavonoids,LF) from the Glycyrrhiza uralensis Fisch. cultivated locally in Ningxia province. Methods: 40 male Sprague-Dawley( SD) rats were randomly assigned into 5 groups as control,model( stress + vehicle,po),fluoxetine group( stress + fluoxetine 10 mg / kg,po) and flavonoids groups( stress + flavonoids 100,300mg/kg,po). A-28-day adult rat chronic unpredictable stress( CUS) model was adopted to elucidate the antidepressant pharmacological effect by forced swimming test( FST),tail suspension test( TST) and open field test( OFT). The hippocampus protein expression level of B cell lymphoma-xl( Bcl-xl) and Cysteine aspartic acid protease-3( Caspase-3) were measured by Western-blotting approaches. Results: The flavonoids can produce the antidepressant effect in CUS model in rat,and the flavonoids can up-regulate the protein expression level of Bcl-xl( P < 0. 01) and down-regulate the the protein of Caspase-3( P < 0. 01) significantly in hippocampus. Conclusion: The antidepressant effect of the licorice flavonoids from Glycyrrhiza uralensis may be related to the protection mechanism of neural apoptosis in hippocampus.
