以对羟基苯腈与硫代乙酰胺为原料,浓盐酸和冰醋酸为溶剂进行化学反应生成对羟基硫代苯甲酰胺,产物不经过纯化,直接在乙醇中与2-氯乙酰乙酸乙酯反应合成非布索坦中间体2-(4-羟基苯基)-4-甲基-噻唑-5-羧酸乙酯。该方法可避免使用硫化氢和高压设备,反应易于处理。
4-hydroxybenzenecarbothioamide was prepared from 4-cyanophenol and thioacetamide using concentrated hydrochloric acid and glacial acetic acid as solvent. The raw product reacted with ethyl-2-chloroacetoacetate in alcohol to get the intermediates of febuxostat,2-(4-hydroxy phenyl)-4-methyl thiazole-5-carboxylate. The improved process avoided the usage of hydrogen sulfide and high pressure equipment,and the reaction was easy to handle.
