目的观察大鼠急性心肌梗死后梗死周边区心肌组织中线粒体融合蛋白2基因(mfn2)和线粒体分裂蛋白(DRP1)表达水平的变化。方法 15只雄性SD大鼠通过前降支动脉结扎法建立大鼠急性心肌梗死模型后,随机分为心肌梗死2 d组,心肌梗死7 d组,心肌梗死14 d组;另设假手术组,每组n=5。利用荧光定量PCR法检测梗死周边区心肌组织中Mfn2和Drp1 mRNA的表达。结果心肌梗死组中Mfn2和Drp1mRNA表达低于假手术组,并在心肌梗死后2周内表达呈逐渐下降的趋势(P〈0.05)。结论线粒体融合和分裂异常导致的能量代谢障碍,可能是心肌梗死后心室重塑及心力衰竭的重要机制之一。
Objective To investigate the altered expression of mfn2 and Drp1 in rats with acute myocardial infarction.Methods The acute myocardial infarction model of 15 male SD Rats were induced by left anterior descending artery ligation,then they were randomly divided into AMI groups(D2,D7,D14).5 male SD rats were sham-operated.The expression of Mfn2 and Drp1 mRNA were detected by real-time PCR.Results Compared to sham group,the expression of Mfn2 and Drp1 of the surrounding tissue decreased in AMI groups,and decreased gradually during the two weeks of post-myocardial infarction.Conclusion The abnormal expression of mitochondrial fusion and fission maybe one of the possible mechanisms of heart failure after AMI.
