目的探讨表皮生长因子(epidermal growth factor,EGF)对小鼠肝外胆管上皮细胞(murine extra-hepatic bile duct epithelial cells,MEBECs)体外培养的影响。方法先采用胰酶和DnaseⅠ,再用Ⅳ型胶原酶消化、分离小鼠肝外胆管组织为单细胞悬液进行接种,接种的细胞分别用含或不含10ng/mL EGF的DMEM/HamsF12(1∶1)培养基进行原代培养。倒置显微镜下观察细胞生长的形态变化,MTT法检测EGF对MEBECs增殖的影响。结果添加EGF组较未添加EGF组MEBECs体外存活时间明显延长(21d±1.3d vs(12.1d±2.1d)(P〈0.001),细胞活力优于未添加组(P〈0.001)。结论培养基中添加EGF可以明显地促进小鼠肝外胆管上皮细胞的增殖,延长细胞体外培养的存活时间。
Objective To explore the influence of epidermal growth factor (EGF) on the growth of murine extrahepatic bile duct epithelial cells (MEBECs) in vitro. MethodsThe extrahepatic bile ducts of BALB/c mice were digested with trypsin in the presence of DNaseⅠand further digested with collagenase Ⅳ. MEBECs were isolated and seeded in a culture flask containing DMEM/Hams F12 (1∶1) supplemented with or without 10 ng/mL epithelial growth factor (EGF) respectively. Then the morphologic changes of the cells were observed with inverted phase contrast microscope and MTT assay detected the influence of EGF on the growth of BECs. ResultsThe duration of the MEBECs of the group with EGF was obviously longer than the other group (12.1±2.1 d vs 21±1.3 d, P 0.001). And the MEBECs of the group with EGF had better cell vigor (P 0.001).ConclusionEGF added to medium can obviously promote the proliferation of MEBECs and prolong the duration of MEBECs in vitro.
