人离体乳腺癌肿瘤新生血管的同步辐射成像实验研究

目的:探讨同步辐射类同轴相衬成像在人乳腺癌离体标本中的应用。方法:12例乳腺癌标本经4%甲醛溶液固定后,在上海同步辐射光源(Shanghai Synchrotron Radiation Facility,SSRF)X线成像与生物医学应用光束线站(BL13W)上进行同步辐射类同轴相位衬度二维及三维成像,最后行病理学及免疫组化检查,将所得影像学资料与病理学资料进行对比研究。结果:物像距为100 cm、能量为13 ke V时,图像具有较高的空间分辨率和衬度分辨率。类同轴相位衬度二维及三维成像均可在无对比剂条件下清晰显示肿瘤新生血管,最小肿瘤血管直径约9μm。结论:同步辐射类同轴相位衬度成像可检测人乳腺癌标本中的肿瘤新生血管,清晰观察肿瘤血管的形态特征和空间分布,为研究肿瘤新生血管的形成机制及各种干预或治疗措施对肿瘤血管的影响提供了检测和监测方法,具有很高的医学应用价值。

英文摘要:

Objective:To evaluate the application of synchrotron radiation (SR) in-line X-ray phase-contrast imaging in human breast cancer.Methods:Twelve excised human breast specimens were studied. Monochromatic SR X-ray in-line phase-contrast images were obtained at the beamline BL13W of Shanghai Synchrotron Radiation Facility (SSRF) in China with the energy of 13 keV and the distances of 100 cm. Meanwhile, the three-dimensional images were processed. Finally, the pathology results were conifrmed.Results:The images obtained with SR had higher contrast and better resolution. The angiogenesis of the breast cancer was obtained for the ifrst time with no contrast agents, and the minimal tumor blood vessel was about 9 μm in diameter.Conclusion:SR in-line X-ray phase-contrast imaging can detect the angiogenesis of human breast cancer, and clearly display the morphological characteristics and spatial distribution of tumor blood vessels. It provides a method to study the mechanism of tumor angiogenesis and the effects of various interventions or treatments on tumor angiogenesis. It would be a great potential diagnostic tool in clinical application in the future.
