目的 探讨上皮间质样转化(epithelial-to-mesenchymal transition,EMT)在鼻息肉(nasal polyposis,NP)中的作用,观察转化生长因子β1(transforming growth factor 31,TGF-β1)对鼻黏膜上皮细胞EMT的影响.方法 留取10例鼻息肉患者及10例作为对照组的鼻中隔偏曲患者的鼻黏膜组织.实时反转录聚合酶链反应(real-time PCR)法检测鼻黏膜组织及原代上皮细胞EMT标志物上皮紧密连接蛋白E-钙黏蛋白(E-cadherin)、β链蛋白(β-catenin)、紧密连接蛋白1(zonula occludens 1,ZO-1)、间质细胞特异性分子波形蛋白(vimentin)、α型平滑肌肌动蛋白(α-smooth muscle actin,α-SMA)mRNA的表达情况,Western blot法检测原代上皮细胞E-cadherin和vimentin蛋白的表达.以SPSS16.0统计软件进行统计学分析.结果 E-cadherin、ZO-1在NP组织中的相对表达量分别为(0.012±0.007)、(0.006±0.003),低于对照组鼻黏膜中的表达,分别为(0.041±0.024)、(0.011±0.005),差异有统计学意义(t=3.675,P<0.01;t =2.956,P<0.05);而NP组与对照组鼻黏膜相比,β-catenin、vimentin和α-SMA的相对表达量差异均无统计学意义(t值分别为0.990、0.429、0.326,P值均>0.05).在对照组鼻黏膜及NP组上皮细胞气液培养体系中,TGF-β1实验组E-cadherin、ZO-1 mRNA表达水平较对照组降低,差异有统计学意义(t对照组分别为3.639、3.430,tNP组分别为3.279、2.864,P值均<0.05),而vimentin、α-SMA较对照组表达水平升高,差异有统计学意义(t对照组分别为-6.393、-3.085,tNP组分别为-2.981、-3.087,P值均<0.05);蛋白质水平:TGF-β1实验组E-cadherin表达水平均较对照组降低,而vimentin表达水平增高,差异有统计学意义(t对照组分别为3.583、-3.844,tNP组分别为5.113、-3.642,P值均<0.05).结论 EMT可能参与了NP的病理生理过程,TGF-β1是引发这一过程的重要因素.
Objective To detect the expression of epithelial-to-mesenchymal transition (EMT) biomarkers in nasal polyposis (NP) and to determine the effect of transforming growth factor β1 (TGF-β1) on EMT in cultured nasal epithelial cells.Methods The specimens were obtained from sinus mucosa of 10 NP patients and inferior turbinate mucosa of 10 nasal septum deviation patients.The difference of mRNA expression of E-cadherin, β-catenin , zonula occludens 1 (ZO-1) , vimentin and α-smooth muscle actin (α-SMA) in tissue and cultured nasal epithelial cells was detected by real-time PCR.The difference of protein exprssion of E-cadherin and vimentin in cultured nasal epithelial cells was detected by Western blot.SPSS 16.0 software was used to analyze the data.Results The relative expression of E-cadherin and ZO-1 in NP tissues (0.012 ± 0.007;0.006 ± 0.003) was higher than in normal nasal mucosa (0.041 ±0.024;0.011 ±0.005), the difference was significant (t =3.675, P 〈0.01;t =2.956, P 〈0.05).However, there was no significant difference in the relative expression of β-catenin, vimentin and α-SMA between two groups (t value was 0.990, 0.429, 0.326, all P 〉 0.05).In cultured nasal epithelial cells both from two groups, TGF-β1 induced the decreased E-cadherin, ZO-1 (t l value was 3.639, 3.430, both P 〈 0.05;tNP value was 3.279, 2.864, both P 〈 0.05) and increased α-SMA, vimentin mRNA expression (tcontrol value was-6.393,-3.085, all P 〈 0.05;tNP value was-2.981,-3.087, both P 〈0.05).Also, TGF-β1 induced the decreased E-cadherin and increased vimentin protein expression (tcontrol value was 3.583,-3.844, both P 〈 0.05;tNP value was 5.113,-3.642, hoth P 〈 0.05).Conclusion EMT is likely to contribute to nasal polyposis and TGF-31 is involved in this process.
