利用牛津杯法筛选到对片球菌素BM-1敏感的植物乳杆菌wQ0815和肠膜明串珠菌05—43。并利用Nisin诱导表达系统,在乳酸乳球菌Nz9000中诱导表达两敏感菌株的甘露糖磷酸转移酶系统IICD组分.抑菌实验表明不同来源IICD组分皆可被片球菌素BM-1识别。进一步将片球菌素BM-1同源免疫蛋白分别与两种IIcD组分同时在乳酸乳球菌中表达,抑菌实验表明:免疫蛋白可识别植物乳杆菌来源的IICD组分.不能识别肠膜明串珠菌来源IICD组分。研究结果为IIa类细菌素的同源免疫蛋白对受体存在特异性识别提供了直接证据。
Lactobacillus plantarum WQ0815 and Leuconostoc mesenteroides 05-43, which are sensitive to pediocin BM-1, were selected by the well-diffusion assay. Man-PTSIICD genes of L. plantamm WQ0815 and L. mesenteroides 05-43 were expressed in Lactococcus lactis NZ9000 by inducible NICE system. The well-diffusion assay indicated that man-PTS IICD of two strains could be recognized by pediocin BM-1. Im- munity protein was respectively co-expressed in L. lactis with man-PTS IICD of two strains. The result showed that immunity protein could recognize man-PTS IICD of L. plantarum WQ0815 but couldn't recognize man-PTS IICD of L. mesenteroides 05-43. These results provided direct evidence for existence of specific recognition between immunity protein and receptor.
