定性观察PGF2α对培养的人眼睫状肌细胞流体阻力的作用,以探讨PGF2α降眼压作用机制。用第4代传代第14天的人眼睫状肌细胞在Transwell上构成流体阻力模型,选取流速差异在15%以内的一对Transwell作为实验对象,观察浓度为2×10^-7mot/L的PGF2α能否使流速较慢的Transwell的流速超过原流速较快者。共做三次并做不加药对照观察。实验组及对照组中流速较慢的Transwell的流速提高均未达到15%。因此浓度为2×10^-7moL/L的PGF2α未能使经过本实验中培养的人眼睫状肌细胞的流体流速提高15%。
The effect of PGF2α to the liquid resistance of cultured human ciliary muscle cells is observed to discuss the mechanism of PGF2α decreasing eye pressure. Liquid resistance model was built with 14 days cultured 4 passage human ciliary muscle cells on Transwell. A pair of Transwell was choosed as experimental objects in which the difference of flow velocity was within 15%. 2 × 10^-7 mot/L PGF2α was added in the Transwell of primary slower flow velocity to observe whether the flow velocity of this Transwell could exceed the another one. Three times were repeated. Control groups without drug were also observed. Flow velocity of the Transwell of primary slower flow velocity did not increase 15% both in experimental and control groups. So 2 × 10^-7 mot/L PGF2α did not increase 15% flow velocity passing through cultured human ciliary muscle cells on Trunswell in this experiment.
