将60只300日龄的蛋鸡随机分成试验组和对照组,每组各30只,分别采用高能低蛋白日粮和正常日粮进行饲养试验,试验期为60 d,于试验的第1、30、60 d随机选取试验组和对照组鸡各6只,应用半定量RT-PCR方法检测了其肝组织中apo AⅠ mRNA的表达情况。结果显示,试验组第30 d和第60 d蛋鸡肝中apo AⅠ mRNA对β-actin mRNA的相对丰度均显著低于同日龄的对照组,特别是在试验第60 d差异达到显著水平(P〈0.05)。在试验组内比较也可以发现,随着试验期的延长,肝中apo AⅠ mRNA的表达量低于前一个试验点,且第60 d时较第30 d时差异显著(P〈0.05)。表明,高产蛋鸡饲喂高能低蛋白饲料一段时间后可导致蛋鸡肝apo AⅠ mRNA的表达量下降。
Sixty layers aged 300 days were randomly divided into experimental group and control group of 30 each. The control group was fed with the standard diet,and the experimental group was fed with the high-energy-low-protein diet for 60 days. The apo A Ⅰ gene and β-actin gene were amplified exactly by semi RT PCR from the total RNA extracted from livers of 6 laying hens picked out randomly from the ex perimental group and the control group on day 1,30 and 60 during experiment, respectively. The apo A Ⅰ mRNA expression in liver of the experimental group decreased compared with that in the control group on days 30 and 60(P〈0.05). apo A Ⅰ mRNA expression on day 60 decreased significantly compared with that in the experimental group on day 30(P〈0.05). The results indicated that prolonged feeding with highenergy-low-protein diet should be responsibility for the decrease of apo A Ⅰ mRNA expression in liver of the laying hens.
