水芹总酚酸对D-半乳糖胺盐酸盐诱导肝损伤的保护作用

中文摘要:

目的观察水芹总酚酸(OJTPA)对D-半乳糖胺盐酸盐(D-Galn)致小鼠急性肝损伤的保护作用。方法将小鼠随机分为生理氯化钠溶液(NS)组、D-Galn模型组、OJTPA低、中、高(125、250、500mg·kg^-1)剂量组和阳性药联苯双酯(LB,150mg·kg^-1)组,除NS组、D-Galn模型组给予与OJTPA高剂量同等剂量的NS外,给药各组均分别给药7d后,以D-Galn(800mg·kg^-1)给各组小鼠(NS组除外)腹腔注射,使之中毒,造成急性肝损伤,24h后杀鼠取材,分别检测各组动物肝功能等指标并取肝脏作病理组织学检查。结果D-Galn模型组小鼠肝功能明显异常,ALT、AST显著升高,肝组织变性、坏死严重。OJTPA中、高剂量组小鼠血清中ALT、AST的水平均非常显著低于模型组(P〈0.001),并呈良好的剂量依赖性;此外,还可显著降低肝损伤小鼠血清丙二醛的含量,显著升高其超氧化物歧化酶活力;肝组织镜检证实,OJTPA中、高剂量均能减轻中毒小鼠肝细胞损伤程度。结论OJTPA对D-Galn致小鼠急性肝损伤有明显的保护作用。

英文摘要:

Aim To observe the protective effects of Total Phenolics Acid of Oenanthe Javanica (OJTPA)on acute liver injury induced by D-Galactosamine HCl in mice.Methods The mice were randomly divided into the NS group,D-Galn group,OJTPA high,middle,low(500,250,125mg · kg^-1 )dosage group and bifendate group (LB,150mg · kg^-1 ) randomly.All groups were given medicine except the NS group which was given the same dosage as the D-Galn model group and the OJTPA high dosage group.Seven days latter,D-Galn (800mg · kg^-1 ) was given to each group of mice (except for the NS group) by intraperitoneal injection to poison them,which induced acute liver injury.24 hours later,levels of liver function were measured in each group respectively,and livers from each group were examined by histopathology.Results The liver function was obviously abnormal in the mice of D-Galn model group.The levels of ALT and AST rose sharply and hepatic tissue was severely degenerative and necrotic.The levels of ALT and AST in the blood serum of mice in the high dosage group of OJTPA were both well below those of the model group ( P 0.001).Moreover,it could also lessen the content of the MDA in the blood serum of injured mice and raised the vigor of SOD.Proved by the microscopic examination of hepatic tissue:a high dosage of OJTPA could lessen the injury of hepatic cells in poisoned mice.Conclusion OJTPA extract can work well to protect from acute hepatic injury induced by D-Galn in mice.
