目的观察BAMBI在非小细胞肺癌(NSCLC)细胞系中的表达分布情况,并探讨其表达与NSCLC病理分型及侵袭潜能之间的关系。方法利用免疫荧光技术检测人肺腺癌细胞系A549以及人肺巨细胞癌具有不同侵袭潜能的亚系BE1、LH7中BAMBI的分布,采用逆转录聚合酶链反应及Western印迹法检测3种细胞中BAMBI的mRNA及蛋白表达。结果BAMBI mRNA在A549细胞系中的表达(0.574±0.035)显著低于BE1细胞系(O.731±0.051)及LH7细胞系(O.734±0.049,P值均〈O.001),后两者间的差异无统计学意义(P=0.216)。A549细胞系中BAMBI蛋白相对含量(0.769±0.103)显著低于BE1细胞系(0.963±0.061)及LH7细胞系(0.957±0.048,P值均〈O.001),后两者间的差异无统计学意义(P=0.096)。BAMBI在A549细胞膜上的染色强度较弱,且呈现颗粒状不均一性;在BE1、LH7细胞膜上的染色较强,连续且均匀一致。结论BAMBI在NSCLC中的表达定位于细胞膜上以及靠近细胞膜的细胞质中,其在肺巨细胞癌细胞中的表达高于肺腺癌细胞,且其表达高低可能与肺巨细胞癌细胞的侵袭潜能无关。
Objective To observe the expression of bone morphogenetic protein and activin membrane- bound inhibitor (BAMBI) in non-small-cell lung cancer (NSCLC) cells and to discuss its relationship with the pathological typing and invasion potentiality of NSCLC. Methods The expression of BAMBI in 3 NSCLC cell lines, including human lung adenocarcinoma cell line A549, one giant cell lung cancer line with high invasiveness potential-BE1 and one with low invasive potential-LH7, were examined by immunofluorescence technology; reverse transcriptase polymerase chain reaction (RT-PCR) and Western blotting were used to determine the expression of BAMBI mRNA and protein. Results BAMBI mRNA expression in the A549 cells was significantly lower than those in BE1 and LH7 (0. 574± 0. 035 vs 0. 731 ± 0. 051, 0. 734± 0. 049, both P〈 0. 001 ) ; there was no significant difference between the latter two (P = 0. 216). The relative amount of BAMBI protein in BE1, LH7 and A549 were 0.769±0. 103, 0.963± 0.061 and 0.957± 0.048, respectively. BAMBI protein expression in A549 cells was significantly lower than that in BE1 and LH7 cells (P〈0. 001 ), and there were no significant differences between BE1 and LH7 cells (P = 0. 096). The BAMBI expression in A549 cells membrane was weak and was in particles, not homogenous; BAMBI expression in BE1 and LH7 membrane was strong, continuous and homogeneous. Conclusion BAMBI protein mainly locates on the membrane and the cytoplasm close to the membrane. The expression of BAMBI in adenocarcinoma of lung is lower than that in giant cell lung carcinoma, and the expression of BAMBI is not related to invasion potential of giant cell lung carcinoma.
