目的研究视黄酸(RA)对心肌细胞Tbx1基因表达的影响。方法分离培养乳鼠心肌细胞,经不同浓度的RA处理后,采用RT-PCR和Western blot方法分别检测乳鼠心肌细胞Tbx1基因在mRNA和蛋白水平表达的变化。结果Tbx1基因在乳鼠心肌细胞有少量基础的表达,在予以RA刺激后,1×10^-7mol/LRA使细胞内Tbx1基因mRNA和蛋白含量表达分别增加1.51倍和1.10倍(P均〈0.05),1×10^-6mol/LRA使细胞内Tbx1基因mRNA和蛋白含量表达分别增加2.21倍和2.38倍(P均〈0.05)。结论RA可以上调乳鼠心肌细胞Tbx1的表达,且此作用呈剂量依赖效应。
Objective To study the regulatory mechanism of retinoic acid (RA) on the Tbxl expression in eardiomyoeytes. Methods Ventricular cardiomyocytes were isolated from neonatal rats and cultured, then treated with different concentrations of retinoic acid. The expression level of Tbxl mRNA and protein were measured by using RT-PCR and Western blot technique respectively. Results Basal expression of Thxl in the control group were determined. In comparison with the control group, the expression level of Tbxl mRNA and protein increased 1.51-fold (q = 104.79, P 〈 0.05) and 1.10-fold (q = 91.38, P 〈 0.05) after being treated by 1×10^-7 mol/L RA, and inreased 2.21-fold (q = 153.76, P 〈 0.05) and 2.38-fold (q = 196.77, P 〈 0.05), repeetively under 1 × 10^-6 moL/L RA treating. Conclusions Retinoic acid could induce the expression of Tbxl in neonatal rat cardiomyocytes and its effect is dose dependent.
