Objective： To establish a PITC pre-column derivation and HPLC-DAD method for the determination of free and hydrolysis amino acids in Motherwort fructus. Methods： The free amino acids were extracted by ultrasonic and the protein was hydrolyzed by 6mol/L of hydrochloric acid. Phenyl isothiocyanate （PITC） was the pre-column derivating reagent, and the content of amino acids was determined with HPLC method. Results： Separation of 18 kinds of amino acids was good, the blank had no interference, and the linear relationship was good. The RSD of precision was between 0.25% and 1.38%, the repeatability was between 0.47% and 1.g6%, the stability was between 0.32% and 1.68%, the sample recovery rate was between 95.48% and 101.48%, and the RSD was less than 5%. The content of free amino acid and hydrolytic amino acid was 0.4525% and 10.0293% respectively. Conclusion： The method could be used to test the content of free amino acid and hydrolytic amino acid accurately, simply and readily, which could provide a theoretical basis for the quality control of Motherwort fructus.
