目的:建立PITC柱前衍生HPLC法同时测定茺蔚子药材中游离和水解氨基酸含量的方法。方法:采用超声法提取游离氨基酸,6mol/L盐酸水解茺蔚子蛋白,以PITC为柱前衍生试剂,采用HPLC法测定茺蔚子药材中氨基酸的峰面积并计算其含量。结果:18种氨基酸的分离良好,空白无干扰,线性关系良好,精密度RSD在0.25%-1.38%之间,重复性RSD在0.47%-1.86%之间,稳定性RSD在0.32%-1.68%之间,平均回收率在95.48%-101.48%之间且RSD均小于5%,游离和水解氨基酸的含量分别为0.4525%、10.0293%。结论:该方法简便、快捷,能准确地测定茺蔚子中游离和水解氨基酸的含量,为茺蔚子的质量控制提供了理论依据。
Objective: To establish a PITC pre-column derivation and HPLC-DAD method for the determination of free and hydrolysis amino acids in Motherwort fructus. Methods: The free amino acids were extracted by ultrasonic and the protein was hydrolyzed by 6mol/L of hydrochloric acid. Phenyl isothiocyanate (PITC) was the pre-column derivating reagent, and the content of amino acids was determined with HPLC method. Results: Separation of 18 kinds of amino acids was good, the blank had no interference, and the linear relationship was good. The RSD of precision was between 0.25% and 1.38%, the repeatability was between 0.47% and 1.g6%, the stability was between 0.32% and 1.68%, the sample recovery rate was between 95.48% and 101.48%, and the RSD was less than 5%. The content of free amino acid and hydrolytic amino acid was 0.4525% and 10.0293% respectively. Conclusion: The method could be used to test the content of free amino acid and hydrolytic amino acid accurately, simply and readily, which could provide a theoretical basis for the quality control of Motherwort fructus.
