考察了鞘氨醇单胞菌DP58对吩嗪-1-羧酸(phenazine-1-carboxylic acid,PCA)的降解特性。以PCA为唯一碳氮源配成无机盐培养基,通过改变PCA的初始浓度和不同的细菌接种量,发现PCA被降解时减少的浓度与细菌细胞数量的增量呈线性关系。通过正交试验设计,以不同温度、pH值、摇床转速的组合确定影响该菌株降解PCA的主要因素为pH值,其次是温度。为分析PCA基团结构与降解的关系,研究了该菌株对PCA的结构类似物吩嗪的降解,结果表明鞘氨醇单胞菌DP58降解吩嗪的能力有限。这些实验结果对今后进一步研究降解途径,从而采取措施抑制PCA降解,有十分重要的意义。
In this study of degradation characteristics of phenazine-1-carboxylic acid (PCA) by the strain Sphingomonas sp. DP58, the PCA added into the inorganic salt culture medium is the sole carbon and nitrogen source of the medium for the strain culture. Experiments show that the PCA concentration decrement caused by degradation is linearly proportional to the increase of cells number of the strain. Using orthogonal experiment, it was found that, among the three main factors affecting the degradation (temperature, pH value and shaker's rotation speed), pH value is the first key factor and temperature is the second one. Moreover, since the phenazine is structurally similar to PCA, the degradation of phenazine by strain Sphingomonas sp. DP58 was studied for researching the relationship between the molecular structure of PCA and PCA degradation. The results show that it is much more difficult for strain Spingomonas sp. DP58 to degrade phenazine than PCA. All the obtained results in this study are significant for further research on the biodegrading pathway to inhibit the PCA biodegradation.
