目的探讨脑梗死后室管膜下区及海马齿状回转化生长因子-β(TGF-β)及血管内皮生长因子(VEGF)的表达及其内源性神经保护作用。方法剖解22例脑梗死后不同时间死亡患者的脑组织,分别从梗死侧的侧脑室室管膜下区及海马齿状回取材(避开梗死灶坏死脑组织),以5例正常脑组织相应部位的标本作为对照组。采用HE染色及免疫组化技术观察不同时间点室管膜下区及海马齿状回TGF-β及VEGF表达的变化及其规律。实验结果应用SPSS13.0软件进行统计学分析。结果 (1)TGF-β阳性细胞于缺血4.5~10h开始增多〔(2.36±0.3)个/HP〕,缺血24~70h达高峰〔(9.1±1.5)个/HP〕,72~96h之后开始下降〔(4.6±1.7)个/HP〕,至缺血120~144h仍有较多阳性细胞〔(1.8±0.3)个/HP〕。对照组有少量TGF-β阳性细胞表达〔(1.1±0.2)个/HP〕;(2)VEGF阳性细胞于缺血4.5~10h开始增多〔(20.9±9.0)个/HP〕,缺血24~70h达高峰〔(48.9±3.4)个/HP〕,72~96h之后开始下降〔(38.5±6.2)个/HP〕,至缺血120~144h仍有少量阳性细胞〔(16.1±6.0)个/HP〕。对照组无VEGF阳性细胞表达。结论脑梗死后,室管膜下区及海马齿状回的神经元及胶质细胞TGF-β及VEGF表达增高,并且二者在表达时间上具有相关性,对脑梗死后神经损伤可能具有协同保护作用。
Objective To investigate the expression and the endogenous neuroprotection of transforming growth factorβ (TGF-β)and vascular endothelial growth factor(VEGF) in the subependymal region and dentate gyrus after human brain infarction.Methods Brain tissues were taken out from ubependymal region and dentate gyrus in 22 dead who died at different time points after brain infarction.Five cases of normal brain tissue samples on corresponding parts were taken as control group.The changes and regularity of TGF-β and VEGF expressions at different time points were observed by HE staining and immunohistochemical method.Experimental results were analyzed by SPSS 13.0 software.Results (1)TGF-βpositive cells started increasing at hours 4.5~10 of ischemia〔(2.6±0.3)cells/HP〕,reaching peak at hours 24~70〔(9.1±1.5)cells/HP〕,starting falling at hours 72~96〔(4.6±1.7)cells/HP〕,a lot of positive cells remaining at hours 120~144〔(1.8±0.3)cells/HP〕.A few TGF-βpositive cell expressions were noted in control group〔(1.1±0.2)cells/HP〕.(2)VEGF positive cells started increasing at hours 4.5~10 of ischemia〔(20.9±9.0)cells/HP〕,reaching the peak at hours 24~70〔(48.9±3.4)cells/HP〕,starting falling at hours 72~96〔(38.5±6.2)cells/HP〕,a few positive cells remaining at hours 120 ~144〔(16.1±6.0)cells/HP〕.No VEGF positive cell expressions were noted in control group.Conclusion After brain infarction,the expressions of TGF-βand VEGF in subependymal region and dentate gyrus increase,and there is correlation in expression time between the two,which is of protection for nerve damage after brain infarction.
